Eucommia ulmoides Oliv. is Eucommiaceae and Eucommia genus,which also known as Sixian,Sizhong and Mumian.It is speciality precious Chinese herbal medicines.Bark and leaves are used for medicine,which could nourish liver and kidney, strengthen bones and muscles,the role of tocolysis.Therefore,it is more widely for Eucommia ulmoides Oliv. as feed additives in animal husbandry to improve the quality of meat and egg.lt can cultivate plants as a nutritional raw materials.On the basis of analyzes and summarizes the chemical constituents and pharmacological effects of Eucommia leaf and Eucommia male flower tea,the HPLC method is used to determinate contents of cholesterol and collagen in eggs and chicken which feed the Eucommia feed additives. The purpose is to examine the effects on the quality of eggs and chicken and chicken skin, and the effect on contents of ginsenoside and amino acid in which Ginseng is cultivated by a nutritional raw materials of Eucommia ulmoides Oliv.And formulation quality standards of Eucommia male flower tea.This will provide a detailed theoretical basis for comprehensive development and utilization of the Eucommia and Eucommia male flower tea.The main results as follows:1.Determination the cholesterol in eggs by RP-HPLC method.The chromatographic conditions were: chromatographic column:Thermo Hypersil Gold C18(250mm×4.6mm,5μm),mobile phase:acetonitrile-isopropyl alcohol was85:15, detection wavelength was206nm,column temperature was15℃,the flow speed was1.0mL/min.Under this condition the results show that cholesterol can achieve the baseline separation from non-measured ingredients and methodology meet the requirements.The results showed that there are some differences on the content of the cholesterol in different species of eggs. the content of the cholesterol is higher in rural grass eggs and the Eggs.the cholesterol is lower in Eucommia eggs,pearls eggs and YuanFang rural grass eggs, these may be related to the cholesterol content of different varieties of chicken feed and the absorption and metabolism of the different varieties of chickens and other factors.2.Determination the hydroxyproline in chicken and chicken skin by ELSD-HPLC method.The chromatographic conditions were:chromatographic Column:Thermo Hypersil the Gold C18(250mm×4.6mm,5μm);mobile phase:0.05%acetic acid solution-acetonitrile (96:4),column temperature was15℃;the flow speed was0.1mL/min,the ELSD parameters:the drift tube temperature was40℃,the carrier gas pressure was0.29MPa.The results show that Eucommia leaves as feed additives have a certain impact on the quality of chicken and chicken skin. It had no significant effect on collagen in chicken,and a greater impact in chicken skin.And there is a correlation between the amount of eucommia leaf and the content of collagen in chicken skin.This showed that Eucommia leaves as feed additives can increase the collagen in chicken skin.3. Determination the ginsenoside in Ginseng by RP-HPLC,and determination amino acid in Ginseng by the amino acid analyzer.And studied on the effect of content of amino acid in ginsenoside when Eucommia leaf as organic fertilize.The results showed:using Eucommia leaf as organic fertilize has little impacted on the content of ginsenosides in ginseng;but has certain impacted on amino acid,which has larger influence on the content of aspartic acid,glutamic acid,arginine,histidine.4. Formulation the quality standards of Eucommia male flower tea. Established the method of microscopic identification of Eucommia male flower tea, it can seen many pollen grains under the optical microscope,which were spherical or blunt triangle,and part of pollen grains wrinkled.Epidermal cell are polygonal in surface view,arranged closely, and there are irregular and dense horny texture.Stoma are infinitive which are oval or oblong, diameter is40μm,longth is50μm,subsidiary cells are four to eight.Catheter mainly is ringed catheter,and also can seen threaded catheter.Rubber showed strips or distorted into a group. It can seen that the diameter of pollen grain is25to35μm with three holes ditch under electronic microscop,and there is the small granular protrusions on the outer.Established the method of TLC identification of chlorogenic acid reference substance.Established the method of HPLC to determination the chlorogenic acid content of Eucommia male flower tea,and carried out the methodology examination to determine the content limit which was0.5%.And investigated Eucommia male flower tea of the microscopic identification, determination of moisture, total ash, acid-insoluble ash and water-soluble extract for the first time, which provide the basis for quality control of Eucommia male flower tea in the future.
