Study On The Effects Of New Veterinary Drug Kuianchun On Immune Function In Mice

Abstract/Summary:
Kuianchun,a new different side chain structural derivative like olaquindox,is a new animal feed additive which was synthesized by Lanzhou institute of animal sciences and veterinary pharmaceutics of Chinese academy of agricultural sciences.The previous study showed that it was less toxicity and residue in edible pork or chicken.In present study.in order to analyze the effects of drug on immune function of the animal and provide scientific data for overall assess clinical safety about kuianchun,we study on immune organ and immunocell effects of kuianchun on mice through immune organ index,splenic lymphocyte transformation,the effect of nitric oxide and nitric oxide synthase,cytokine level in serum.RESULTS: (1) After being continuously administered kuianchun for 7 days and 14 days,spleen index of drug treated group was higher than control group,thymus index was no significant difference among treated group.The percentage of the ANAE-positive cell of male mice was increased,the percentage of the ANAE-positive cell of female mice was decreased.Kuianchun can promote splenic lymphocyte transformation in coordination with ConA at 200μg/ml and 800μg/ml. (2) The effect of kuianchun on the nitric oxide content of spleen and thymus was different.The nitric oxide content of female mice spleen was increased,the nitric oxide content of thymus was decreased. The nitric oxide synthase content of spleen was increased,the content of inducible nitric oxide synthase has no significant difference.Kuianchun can decrease the nitric oxide content which was formed by peritoneal macrophage through LPS induction. (3) The change of cytokine TNF and IL-6 was identical.The content was decreased at high dose and was increased at low dose in male mice serum,the content was decreased when dose was increased in female mice serum. The content of IL-2 is concerned with the sex of mice and the time of administration.CONCLUSIONS: (1) The effect of kuianchun on immune organ index has no significant sex difference.Kuianchun can promote splenic lymphocyte transformation in coordination with ConA. (2) Kuianchun can decrease the nitric oxide content which was formed by peritoneal macrophage through LPS induction. (3) The effect of Kuianchun on the production level of TNF,IL-2,IL-6 in serum of mice may have sex and dose difference.
