The extracting conditions to produce dried immunoglubin from fresh pig blood were screened on this study. By saturated ammonium sulfate fractionation step-by-step. Then the effect of the product on immune function in mice was discussed.In Expt.1, the precipitation effect among the caprylic acid and the saturated ammonium sulfate fractionation were compared according to their precipitated abilities on the basis of the content of IgG. As a result, the precipitation effect of saturated ammonium sulfate fractionation was better than the caprylic acid.In Expt.2, The technological conditions of the saturated ammonium sulfate were discussed. The orthogonal test L9 (34) was used to evaluate the factors optimum conditions based on single factor test. The single factor test optimum conditions of saturated ammonium sulfate fractionation were established as below:①extracting temperature 30℃, on this condition, the content of IgG was maxlmum for 81.75%.②standing time 45 min, on this condition, the content of IgG was maxlmum for 87.43%.③reaction pH 7.0, on this condition, the content of IgG was maxlmum for 81.47%. The orthogonal test L9 (34) optimum conditions of neutral protease were established as below: extracting emperature 20℃, standing time 45 min, reaction pH 7.4. The content of IgG was up to 88.08%.In Expt.3, The study was conducted to investigate the effects of pig blood IgG supplementation on immune function of mice after cyclophosphamide challenge and its immune suppression. Mice were allotted to ten treatments including:①normo-control;②oleanolic acid;③low dose IgG;④middle dose IgG;⑤high dose IgG;⑥normo-control+Cy;⑦oleanolic acid+Cy;⑧low dose IgG+Cy;⑨middle dose IgG+Cy;⑩high dose IgG+Cy; On d 3 and d 6, mice in the model groups were injected intraperitoneally with Cy 75mg/kg BW. Whereas mice in the IgG test groups were intragastric administrated with dose of 45, 90, 180mg/kg BW respectively, mice in the oleanolic acid group was intragastric administrated with dose of 36mg/kg BW, mice in normo-control groups were intragastric administrated with an equivalent dose of sterile saline. At the end of the test, weighed the body weight, and eyes blood samples were collected, then the mice were dissected to collect spleen and thymus for evaluation their index. The level of IgG, albumin, globulin, total protein and immunoglobulin was determined. The results showed that: (1) Cy reduced the weight of spleen and thymus, lower than normo-control group, the different doses of IgG protected conspicuously (P<0.05), and presented dose-effect relation. Low dose IgG was fit for production. (2) Cy reduced the level of DTH, but the different doses of IgG antagonized it, compared with normo-control group the antagonistic effect was significant (P<0.05), the normo-control+Cy group was inconspicuous (P>0.05). (3) Compared with normo-control group, the different doses of IgG could antagonize the reduced phagocytosis rate and index of phagocytosis, but the difference was inconspicuous (P>0.05); compared with model normal control group, the middle IgG could antagonize the reduced phagocytosis rate significantly (P<0.05), but the index of phagocytosis was inconspicuous (P>0.05), there was none dose-effect relation. (4) The different doses of IgG increased the content of IgG in mouse blood-serum, compared with normo-control group the effectiveness was significant (P<0.05), especially, high dose IgG was the most significant; compared with normo-control+Cy group, the content of IgG in mice blood-serum was increased, but the effectiveness was inconspicuous (P>0.05), presented dose-effect relation, which indicated that the IgG could increase the humoral-mediated immunity on some degree, and antagonized the level of IgG in serum by cyclophosphamide significantly (P<0.05). (5) Compared with the normal control group, the level of immunoglobulin in serum significantly enhanced by intragastric administration IgG (P<0.05), but not in a dose-effect relation; compared with the model normal control group, the level of IgG in serum was increased, but the difference was inconspicuous (P>0.05), and not in a dose-effect relation. (6) Compared with the normal control group, the level of globulin in serum was significantly increased by intragastric administration IgG (P<0.05), but not in a dose-effect relation, compared with the model normal group the difference was inconspicuous (P>0.05).In Expt.4, the experimental animal was healthy SPF mice, half male and half female, weight 18±2g, mice were allotted to two treatments including:①normo-control(intragastric administration sterile saline);②IgG experimental group. On the first day of the test, the test group intragastric administration IgG(10g/kgBW), observed the mice for 15 days consecutively, weighed weekly, and recorded death rate. The results show that: body weight of the test group increased significantly, during the experimental stage there was none death. So the test proved that the IgG was non-toxic.
