Aflatoxin B1?AFB1?is immunotoxic,and can inhibit the development of immune organs and the function of immune cells and immunoactive substances,causing immune function suppression.oxidative stress and apoptosis are important pathological mechanisms of it.Lycopene?LYC?,a natural pigment found in solanaceae plants,has antioxidant,anti-tumor,anti-inflammatory and immune modulating function.To investigate the interventional effect and mechanism of LYC on immune function suppression in AFB1-induced spleen of mice,male Kunming mice were selected as subjects.Firstly,setting the concentration gradient of AFB1 and detecting the body weight,spleen weight,spleen coefficient,splenic microscopic structure and cytokine content of mice is to select the dose of AFB1 in the next LYC interventional experiment.Secondly,mice were random Ly divided into four groups: control group?CG?,AFB1 treated group?AG,0.75 mg/kg AFB1?,LYC intervention group?ALG,0.75 mg/kg AFB1 + 5 mg/kg LYC?and LYC control group?LG,5 mg/kg LYC?.The spleen structure and function,oxidative stress state and apoptosis level were observed to evaluate the effect and mechanism of LYC on AFB1-induced immune function suppression.The results were shown as follows:?1?Compared with CG,the body weight,spleen weight and spleen coefficient in AG were significantly decreased?p<0.05;p<0.01?.Compared with AG,the above results in ALG were significantly increased?p<0.05?;In CG and LG,the boundary between the white pulp and the red pulp zone is clear,the structure is complete,tand the arrangement is tight.In AG,the boundary between the white pulp and the red pulp zone is not clear,the red pulp congested,the white pulp lymphocyte was reduced,intercellular gap was enlarged,the cells were loosely arranged.In ALG,the red pulp congestion was alleviated,the white pulp lymphocyte was increased,the cells were tightly arranged,the boundary between the white pulp and the red pulp zone was clear,indicating that LYC alleviates AFB1-induced splenic structure injury.?2?Compared with CG,the CD3+,CD4+ and CD8+ T lymphocytes subsets in spleen of AG mice were significantly decreased?p<0.01?.Compared with AG,the above results in ALG were significantly increased?p<0.05;p<0.01?;Compared with CG,the levels of IL-2,IL-4 and TNF-?in serum of AG mice were significantly decreased?p<0.01?,the m RNA expressions of IL-?,IL-2,IL-4,IL-6,IFN-? and TNF-? in spleen were significantly decreased?p<0.01?;Compared with AG,the level of TNF-? in serum of ALG was significantly increased?p<0.01?,the m RNA expressions of IL-?,IL-2,IL-4,IL-6,IFN-? and TNF-? in spleen were significantly decreased?p<0.05;p<0.01?.The m RNA expressions of IL-?,IL-6 and TNF-? in spleen of LG mice were significantly higher than CG?p<0.05;p<0.01?,indicating that LYC alleviates AFB1-induced inhibition of spleen immune function.?3?Compared with CG,the contents of ROS,H2O2 and MDA in spleen of AG mice were significantly increased?p<0.01?,CAT,SOD activities and T-AOC were significantly decreased?p<0.01?;Compared with AG,ROS,H2O2 and MDA contents in ALG were significantly decreased?p<0.05;p<0.01?,CAT,SOD activities and T-AOC were significantly increased?p <0.05?,indicating that LYC can alleviate AFB1-induced spleen oxidative stress.?4?Compared with CG,apoptosis rate,Cyt-c protein expression,Capsase-3 and Capsase-9m RNA expressions and activities,and Bax m RNA and protein expression in AG mice were significantly increased?p<0.05;p<0.01?,MMP,m RNA and protein expression of Bcl-2 and the ratio of Bcl-2 / Bax were significantly decreased?p<0.05;p<0.01?;Compared with AG,apoptosis rate,Cyt-c protein expression,Capsase-3 and Capsase-9 m RNA expression and activity,and Bax m RNA and protein expression in ALG mice were significantly decreased?p<0.05;p<0.01?,MMP,m RNA and protein expression of Bcl-2 and the ratio of Bcl-2 / Bax was significantly increased?p<0.05;p<0.01?,indicating that LYC can inhibit AFB1-induced apoptosis.Conclusion: LYC can effectively alleviate AFB1-induced spleen immune inhibition,and is related to alleviating oxidative stress and inhibiting apoptosis.
