Objective: Extract isoflavone and saponin from Cicer arietium L. at the same time, then to separate, purify and analyze, appraise them, based on this to validate their decreasing blood glucose, regulating blood lipid effect on diabetic mice, by observing their effect on oxidation and anti-oxidation in diabetic mice to discuss their effect on decreasing the level of blood glucose and regulating the metabolism of blood lipid. Methods: (1) Isoflavone and saponin was isolated from Cicer arietium L. by optimal technics that was established according to their character. By contrasting experiment to choose effective method to eliminate fat and protein. (2) Select proper resin to purify and enrich the content of isoflavone and saponin in Cicer arietium L., based on this to screen out the best way to purifying them and to identify them by their universality, color feedback and UV. (3)Build ideal model of DM2 to validate the reducing blood sugar and regulating the metabolism of blood lipid effect of Cicer arietium L.isoflavone and saponin and then by observing its effect on oxidation status in diabetic mice to study the mechanism of their effect. Results: (1) Using ethanol and circumfluent distillate method can extract isoflavone and saponin from Cicer arietium L. effectively at the same time. (2) The best synchro technics of extracting isoflavone and saponin from Cicer arietium L. was: 10times 70% ethanol, in 70℃, extract 3h for two times. (3) centrifugal method and equipotential method are ideal method to eliminate fat and protein. (4) H103 resin and HPD-600 resin can eliminate impurity and purify, enrich the content of isoflavone and saponin in Cicer arietium L. effectively. (5) Polyacetamide chromatogram method is rather than solvent method and can separate and purify Cicer arietium L. isoflavone and saponin effectively. (6) Using the method that after 4 weeks high sugar and high fat feeding, the mice were injected with 25mg/kg streptozotocin at tail vein can build ideal model of DM2. (7)Cicer arietium L. isoflavone and saponin can decrease the level of blood glucose of diabetic mice and has the function of regulating the metabolism of blood lipid. (8) Cicer arietium L. isoflavone and saponin can improve anti-oxidation ability and clear oxygen free radical. This may be one of the mechanism of the regulating metabolism of blood lipid, decreasing blood glucose functions. Conclusion: Isoflavone and saponin were the main chemical compositions of Cicer arietium L., and improving anti-oxidation ability and clearing oxygen free radical was main cause resulting in regulating metabolism of blood lipid and decreasing blood glucose.
