Objective: Due to intensive farming and feeding of high-energy low-protein feeds,a large amount of lipid deposits in the liver of laying hens is prone to cause fatty liver syndrome,which seriously affects the production and health of the laying hens.In this paper,by adding a certain dose of grape seed proanthocyanidin extract(GSPE)to layer diets,the effect of GSPE on the lipid metabolism,antioxidant and egg production performance of layering hens was explored,and the best application plan was screened out.Materials and methods: A total of 200 330-day-old Hailan brown hens with a healthy body condition,similar weight and egg production rate were selected.Randomly divided into 4 groups,each with 5replicates,and each replicate with 10 chickens.Laying hens in the blank group were fed a basal diet.Respectively,125 mg/kg,250 mg/kg and 500 mg/kg of GSPE were added to the basal diet in the low,medium and high dose groups,pre-feeding for 1 week,and the formal trial for 8 weeks.Explore the effect of GSPE on the performance of laying hens by recording feed intake,egg production and egg weight;Explore the effect of GSPE on egg quality by observing ESI,ES,ET,AH,HU and egg yolk cholesterol;By detecting the changes in serum and liver GSH-Px,CAT,T-AOC,SOD,MDA,AST,ALT,TC,TG,LDL-C and HDL-C and detecting the pathological tissue sections of the liver,explore the presence of GSPE anti-oxidation and lipid metabolism of laying hens;Real-time fluorescent quantitative PCR was used to detect the expression of related genes FXR,FGF19 and ASBT m RNA,and explore the effect of GSPE on lipid metabolism of laying hens.The content of this study is divided into two parts,and the specific test results are as follows:1.The effect of GSPE on the performance of laying hens and egg qualityCompared with the control group,the egg production rate of the high and medium dose groups was significantly increased(P<0.05).The egg weight of each test group had a significant increase(P<0.05).The daily feed intake of the middle-dose group was significantly increased(P<0.05).The feed-to-egg ratio of the high and medium dose groups decreased significantly(P<0.05).The rate of soft egg broken in the experimental group decreased significantly(P<0.05).There was a significant difference in eggshell strength in the low-dose group(P<0.05).The specific gravity of egg yolk in the high and medium dose groups was significantly reduced(P<0.05).Egg yolk cholesterol was significantly reduced in the high-dose group(P<0.05).2.The effect of GSPE on the anti-oxidant and lipid metabolism of laying hens(1)Compared with the control group,the liver index and liver fat rate of the high and medium dose groups were significantly reduced(P<0.05).Compared with the serum of the control group,the AST of the test group had a significantly reduced(P<0.05).The low-dose group effectively reduced the content of ALT(P<0.05).Compared with the liver of the control group,AST and ALT were significantly reduced in the middle-dose group(P<0.05).(2)Antioxidant enzyme activity in serum and liver: Compared with the serum control group,the GSH-Px content of the middle-dose group was significantly increased(P<0.05).CAT in the test group was significantly increased(P<0.05).The MDA of the test group was significantly reduced(P<0.05).T-AOC and SOD had increased significantly in the low-dose group(P<0.05).Compared with the results of the liver control group,GSH-Px,T-AOC and CAT in the high and medium dose groups showed a significant increase(P<0.05).SOD has a significant increasing trend in the high-dose group(P<0.05).The MDA of the test group was significantly reduced(P<0.05).(3)Serum and liver lipid metabolism indicators: Compared with the serum blank group,the low-dose group LDL-C was significantly reduced(P<0.05).HDL-C in the experimental group was significantly increased(P<0.05).Compared with the results of the liver in the blank group,TG was significantly lower in the middle-dose group(P<0.05),and TC in each experimental group was significantly lower(P<0.05).(4)Adding a certain dose of GSPE to the diet can significantly or extremely significantly reduce the expression of FXR m RNA(P<0.05/P<0.01).Adding GSPE can significantly reduce the expression of FGF19 m RNA(P<0.01).Adding medium and low doses of GSPE can significantly reduce the expression of ASBT m RNA(P<0.05).Conclusion: The results showed that adding GSPE to layer diets can improve layer performance,lipid metabolism and antioxidant properties.When the addition amount of GSPE is 250 mg/kg,the improvement effect on laying hens is the best.
