Study On New Drug For Endometritis In Cows

Endometritis is a common reproductive disease of cows which caused mainly by infection of pathogenic microorganism. Endometritis is one of the main reasons of infertilitas feminis and causes great loss of economic and reproduction. Antibiotics are used usually for treatments of endometritis, but neotype medicines for endometritis are needed, as problems of drug resistance of bacteria and medicine residue are outstanding. The aim of this paper is to study and manufacture compound recipe of Chinese herbal and chemical medicines for endometritis of cows.Eight kinds of Chinese herbal medicines such as Forsythia suspense, Coptis chinensis, Salvia miltiorrhiza , Pieplant, Folium isatidis, Leonurus, Angelica and Safflower which are often used for therapy of endometritis in cows were selected and antibacterial effect of them was tested on the main pathogenic bacteria (Staphylococcus aureus , streptococcus,Escherichia coli ) causing endometritis in cows in vitro. The medicines which had better antibacterial action than others such as Forsythia suspense, Coptis chinensis and Pie plant were selected by the experiment of bacteriostasis which also offered reference to studies of antibacterial effect of Chinese herd medicines. Three compound recipes were designed according to the result of bacteriostasis experiment , effect of Chinese herb medicines on uterine smooth muscle and Chinese medical science theory .Compound recipe 1 was composed of Forsythia suspense ,Pieplant,Leonurus and Safflower. Compound recipe 2 was composed of Coptis chinensis, Pieplant, Leonurus and Salvia miltiorrhiza . Compound recipe 3 was composed of Coptis chinensis, Forsythia suspense and Leonurus. The result of bacteriostasis experiment showed that three compound recipes could effectively inhibit bacteria and compound recipe 1 was the best among them. Bacteriostasis experiment of four kinds of antibiotics(Penicillin ,Streptomycin ,Neomycin , Ciprofloxacin )often used in the treatments of endometritis in cows has also been done in this study. Results showed that Neomycin and Ciprofloxacin had very good antibacterial action, while Penicillin and Streptomycin were not sensitive to Escherichia coli and Streptococcus. Desensitize of Penicillin and Streptomycin might be caused by drug resistance of bacteria which was created by long-term use of them. The antibacterial action of Ciprofloxacin was the best among four kinds of antibiotics and its biggest antibacterial diameter was 42mm.The antibiotic sensitive experiment also afforded reference for the selection of clinical medicines.EDM composed of compound recipe 1 and Ciprofloxacin was prepared according to standard of uterine perfusate. Safety of EDM was tested by two experiments. Experiment of acute toxicity showed that EDM was innoxious material (LD₅₀>31250mg/kg). Experiment of eye irritation showed that EDM only had bland stimulus and no cacoethic stimulus to mucous membrane.Sixty-five cows with different types of endometritis were treated with EDM and Ciprofloxacin in order to study clinical effect of EDM.
