Cow endometritis is one of the important causes of reproductive disorders in dairy cows.It often causes symptoms such as prolonged estrous cycle and difficult conception,which seriously affects the economic benefits of dairy farming.Therefore,this experiment established a traditional Chinese medicine compound to treat cow endometritis.Firstly,the treatment mechanism of traditional Chinese medicine compound prescription is preliminarily predicted by using network science.And the treatment effect was improved by fermentation,which was verified by in vitro test.Finally,the clinical trial proved the therapeutic effect of the traditional Chinese medicine compound on cow endometritis.The purpose is to provide reference and scientific basis for the traditional Chinese medicine compound for the treatment of cow endometritis,so as to reduce the frequent use of antibiotics.First of all,by checking the literature and "Chinese Veterinary Pharmacopoeia",formulating the prescriptions according to the dose of the drugs and the combined effect of each other,and finally determining the traditional Chinese medicine prescriptions as Salvia,Motherwort,Scutellaria,and Forsythia.Through network pharmacology,the potential targets of effective components of compound Chinese medicine in the treatment of cow endometritis were studied,and the mechanism of the target pathway was expounded.The TCMSP database was used to screen the target of drug components,and 133 active ingredients were screened out from the traditional Chinese medicine compound,mainly including quercetin,kaempferol,baicalein,tanshinone IIA,and 292 corresponding target proteins were found through the drug components.Through NCBI,Gene Card,gene Map,TTD,and Drug Bank,the related targets of dairy cow endometritis were searched,and 130 related targets of dairy cow endometritis were found,and the drug component targets and cow endometritis related targets were screened out.There are24 gene targets in total,which are used as the predictive targets of the Chinese herbal compound for the treatment of endometritis in dairy cows and uploaded to a string database to establish protein interactions(PPI)network,and screened out 7 core targets,including tumor necrosis factor,protein kinase,prostaglandin endoperoxide synthase 2,CSC1-like protein,cellular tumor antigen,Interleukin-1β and interleukin-6.Metascape database was used to analyze the GO function and KEGG pathway enrichment of core targets.Among all the gene pathways of the analyzed core targets,20 are involved in biological processes,6 are cell components,and 6 are molecular functions.The pathways of biological processes are mainly positive regulation of leukocyte-cell adhesion,lipopolysaccharide response,Inorganic response,cellular response to chemical stress,fever response,response to IL-1,regulation of inflammatory response,Positive regulation of tumor necrosis factor,protein phosphorylation,leukocyte and vascular endothelial cell adhesion regulation,rigidity,cytokine-mediated signaling in glandular development,tissue remodeling,development of reproductive structures,development of the digestive system,regulation of epithelial cell migration,response to viruses,positive regulation of nucleocytoplasmic transport,and regulation of stem cell differentiation.Cell components show that the target function mainly acts on cell membrane rafts,membrane side,granule secretory cavity,cytoplasmic perinuclear region,transcriptional regulatory complex,collagen extracellular matrix;molecular functions mainly include signal receptor activity regulation,transcription coactivator protein binding,heme binding,integrin binding,phosphatase binding,regulation of protein homodimerization activity.The KEGG pathway enrichment analysis involves 20 genes,and 9 pathways are involved in regulation.Among them,the genes involved more include AGE-RAGE signaling pathway,cancer proteoglycan pathway,blood flow shear stress and atherosclerosis pathway and Th17 cell differentiation pathway.Finally,the therapeutic effect of TCM compound was determined,which provided theoretical basis for subsequent experiments.Based on the increase of flavonoids and polysaccharides,the effective active ingredients of traditional Chinese medicine.After fermentation,the strain of traditional Chinese medicine fermentation was screened and identified,the fermentation conditions of traditional Chinese medicine were optimized by response surface analysis.The final results showed that Lactobacillus rhamnosus had the best fermentation effect on traditional Chinese medicines.The best fermentation conditions for traditional Chinese medicines were fermented for 6 days at a fermentation temperature of 37 ℃ and a bacterial inoculation amount of 5% for 6 days.The highest content of flavonoids in the experiment was 25.37%.And results showed that the minimum inhibitory concentration of traditional Chinese medicine increased by 25%.The drug safety test was carried out in mice.In the drug sensitization test and uterine perfusion test,the mice showed no abnormality,which proved that the drug was safe and met the clinical medication standards.The results of clinical trials show that the compound fermented traditional Chinese medicine can reduce the number of neutrophils in the uterus of dairy cows,thereby alleviating the inflammatory symptoms of endometritis.It can be seen from the B-ultrasound image that the drug can promote uterine contractions through drug components.Reduces the accumulation of purulent fluid in the uterus.It has a benign effect on cow endometritis.
