Studies On The Chinese Herbal Medicine-Gongdejian For Treatment Of Endometritis In Dairy Cows

The Coptis chinenisis Franch,Angelica sinensis (Oliv.) Diels,Carthamus tinctorius L,Leonurus japonicus Houtt and Mentha-camphor were selected as main componets to compose Gongdejian-an uterus lavage solution to treat the endometritis in dairy cows, the composition of Gongdejian was based on the modern research materials of Chinese herbal medicine and effectively medical prescription. The stability,toxicity,security,anti-inflammatory effect,minimal inhibitory concentration in vitro,effect of meidication in clinical cases of Gongdejian were studied.In the stability test, when exposed to the 2000~4000lx illumination for 10 days, the appearance character,pH and the absorbance between 200~400nm had no notable changes, conforming to quality criteria; Under the condition of 40±1℃,75% of relative humidity, the appearance,pH and the absorbance between 200~400nm had no significant changes, consistent with quality criteria. The results showed that Gongdejian was stable both to strong light and high temperature.Giving the maximum concentration and the maximum capacity to mice by intragastric administration, the LD50 couldn't be measured, indicating that Gongdejian was low of acute toxicity. In the maximum tolerable dose (MTD) experiment, the results showed MTD>40g/㎏; In the long-term toxicity test, high dosage group and low dosage group were set up. The intragastric administration dose were 4,0.4ml/100g body weight of rats (for 200 times and 20 times of clinical dose) per day for 8 weeks, the results showed there were no significant changes in the rats's behavior and activity, the body weights, the organs viscera coefficient, the indexes of hemotology and blood biochemistry compared with the control group. No pathological changes were detected in most organs, indicating that long-term administration of Gongdejian was non-toxic.In the haemolytic test, the RBC of rabbits were hemolyzed by Gongdejian, as a result, Gongdejian could not be injected intravenously; In general allergy test, Gongdejian caused no allergic reaction to cavy; In the irritanting test to eyeballs and vaginal mucosa irritant test, there were only slight hyperemia to the conjunctiva, which returned normal within 60 minutes, and there were no obvious histopathological changes in the vaginal mucosa of cavy. The results showed that Gongdejian had no irritant effect and was safty for uterus lavage.The result of antiinflammatory test indicated that the high dosage group (20g herb/kg body weight) and the middle dosage group (10g herb/kg body weight) could obviously inhibit (P<0.05) the swelling of mice's ears which was caused by xylene, as well as the accrementition of granulation tissue caused by cotton balls. The antibactery test in vitro showed that the minimum inbibitory concentration (MIC) of Gongdejian to the E coli,Staphylococcus,Pseudomonas and Streptococcus were 0.0125,0.1,0.25,and 0.25g herb /ml, respectively.Uterine secretion was collected aseptically from 12 dairy cows with endometritis, after fractional cultivation and biochemistrical identify, 8 strains of E coli (42%),6 strains of Streptococcus (32%),4 strains of Staphylococcus (21%) and 1 strain of Pseudomonas(5%) were detected. 5 (42%) cows were infected by single bacteria, 7 (58%) cows by two species bacteria. 4 and 8 cows that were respectively 5~10 days and 11~20 days after parturition were treated with Gongdejian. The cure rate and the pregnancy rate at first insemination were 100%, 75%,and 87%, 63%, respectively. The cure rate and the pregnancy rate of all cows were 92% and 67%.
