| With the continuous expansion of beef cattle farming,the incidence of endometritis is increasing year by year.At present,the drugs for the treatment of endometritis in beef cattle are mainly antibiotics,but the long-term use and abuse of antibiotics have led to a gradual increase in drug-resistant strains and poor clinical effects.In this study,the principles of antipyretic and analgesic,blood circulation and stasis removal were adopted,and traditional Chinese medicines such as angelica,motherwort,Chuanxiong,Shengdi,red peony,and ze diarrhea were selected to form self-formulated compounds.The clinical efficacy and mechanism of action of this traditional Chinese medicine compound for endometritis in beef cattle were studied,which provided a reference for the further development of low-toxicity,residue-free and non-resistant traditional Chinese medicine compound for the prevention and treatment of this disease.This study had the following components:1.20 beef cows with endometritis were randomly divided into control group,low-dose treatment group,medium-dose treatment group and high-dose treatment group,with 5 cows in each group.Each cow in the low,medium and high dose treatment groups was given 0.4 kg,0.5 kg and 0.6 kg of the self-prepared Chinese herbal formula once a day for 14 d.The clinical treatment effect was counted on the 15th day,and two blood samples were collected before morning feeding,one for determining the ratio of peripheral blood T lymphocyte subpopulation,lymphocyte transformation rate and apoptosis rate.One blood was used to determine the percentage of peripheral blood T-lymphocyte subpopulation,lymphocyte transformation rate and apoptosis rate.The results showed that different doses of Chinese herbal compound had good efficacy on clinical endometritis in beef cattle;compared with the control group,the serum CD4+ level was significantly higher(P<0.05),CD4+/CD8+was highly significant(P<0.01),IL-2,IL-4 and IL-6 levels were on the rise,and IL-4 and IL-6 levels in the middle and high dose groups(P<0.01),IgM and IgA levels were significantly increased in the middle and high dose groups(P<0.05),and the levels of AMH,E2 and INHB were highly significant(P<0.01).2.The UPLC-MS/MS metabolomics technique was used to analyze the modulating effects of the herbal compound on serum metabolites in beef cattle with endometritis.28 differential metabolites were screened(P<0.05,VIP ≥1,FC>2 or FC<0.5),of which 20 were up-regulated and 8 were down-regulated.The enrichment of the pathways showed that the self-prepared herbal compound could exert anti-inflammatory and reproductive performance enhancing effects by up-regulating tyrosine,a key metabolite in the biosynthetic metabolic pathway of phenylalanine,tyrosine and tryptophan,folate,a key metabolite in the folate metabolic pathway and biotin,a key metabolite in the biotin metabolic pathway.In conclusion,different doses of the self-prepared herbal compound had good efficacy on endometritis in beef cattle,and the middle and high dose groups could significantly improve the immune function and reproductive hormone levels in beef cattle;the herbal compound achieved the effects of treating endometritis and improving reproductive performance in beef cattle by up-regulating the levels of tyrosine,folic acid and biotin. |
