The experiments were carried out on the farm,lab of USTC and five provinces of china from 2004 to 2006,by using the principle and methods of tobacco chemistry,biochemistry,tobacco cultivation,tobacco biochemistry. Mainly has studied about the change of the polyphenol and the correlation chemical substance in leaf tobacco in the different vegetal period, the different rank and in the different ecology area. The main results are as follows:1.With the growth and development of tobacco leaf, the content of total phenols,chlorogenic acid and rutin increased gradually.The content of Phenylalanine and tyrosine(the Precursor of polyphenols) decreased all the time in tobacco leaf during the growth. It was significantly negative correlation between total phenols and phenylalanine,tyrosine during the growth of tobacco plants. It was positive correlation between PAL activity and total phenols,chlorogenic acid,rutin. And it is close to significant correlation level.No correlation was found between PPO activity and ployphenols contents. The difference between total phenols and nicotine was significant,and total phenols had negative correlation with total nitrogen. The difference between rutin content and total nitrogen was not significant,and the other relations are the same as total phenols.2.Along with elevating, the polyphenols matter, the nicotine, the total nitrogen content all elevates in the flue-cured tobacco, the sugar to nicotine compares drops. But the sugar content is highest in middle spot, upside is next, lower part is lowest. In the same spot, along with leaf tobacco color deepening, the polyphenol, the sugar content reduces, but the nicotine and the total nitrogen content actually increases, the sugar to nicotine drops. When spot and color are all same, along with the rank enhancement, various chemical substance content change rule is extremely obvious. polyphenol, nicotine, the total sugar and the original state sugar content rises, but the total nitrogen content is drops.3.The chemical compositions have difference between different regions, and the difference is a little greater in some areas in the same condition.Nicotine , and sugar/nicotine has significant difference between regions,the difference is less for total nitrogen ,the difference of the absolute value of sugar is greater,but the relative value is less.The characteristics of the chemical composition of all producing regions are that:nicotine content in Liaoning is the lowest, sugar content is in the middle, total nitrogen content and the ratio of sugar to nicotine content is medial. In the tobacco leaves in Anhui,nicotine content is the greatest, total sugar content, sugar/nicotine is the lest,reductive sugar content is lower,total nitrogen is higher. In the leaves in Guizhou,nicotine content is higher, total nitrogen content is medial, sugar/nicotine is high. In Fujian, total sugar and reductive content is medial or high,total nitrogen is medial, sugar/nicotine is higher.In Yunnan,nicotine,total sugar ,reductive content is higher, sugar/nicotine is medial or lower, total nitrogen is the highest. By the comparison of both the south and north producing region,In the north,sugar conten, sugar/nicotine is low;Nicotine and total nitrogen content is high;In the south,sugar content and sugar/nicotine is high,Nicotine and total nitrogen conteni is low.
