ObjectiveThis study with estradiol benzoate and oxytocin under the condition of low temperature freezing haemorrheological nature due to the type of dysmenorrhea rat animal model as the research object, through the observation of moxibustion cell palace related acupuncture point (Sanyinjiao(SP6), Guanyuan(RN4)group) for this model, the influence of body torsion reaction and midbrain κ、μ receptors predominate in the rat spinal cord changes as observation indexes, to preliminarily moxibustion to alleviate the central role of primary dysmenorrhea mechanism, at the same time explore the difference of the analgesic effect between Sanyinjiao(SP6) and Guanyuan(RN4).Methods40 SD female rats (in estrogen between the stage) is divided into:saline group, haemorrheological nature class dysmenorrhea, Sanyinjiao(SP6) and Guanyuan(RN4)group by the random number table method,10 in each group. Saline group daily to give isotonic saline treatment, not a bit; Haemorrheological nature card type dysmenorrhea group rats given subcutaneous injections of estradiol benzoate, daily for 10 days, the last for 1 h after intraperitoneal injection of oxytocin, and 1-5 d after injecting estradiol benzoate into the daily-25 ℃ freezer frozen 4 hours, opened the ventilation stopover in 2 hours 5 seconds, the preparation of haemorrheological nature card type dysmenorrhea rat model, not a bit; Sanyinjiao(SP6) group and Guanyuan(RN4)group building methods the same to haemorrheological nature card type dysmenorrhea group, and in building 6,8,10 days to moxibustion after the treatment, each successive moxibustion three strong. Observed within 20 min after injection of oxytocin body torsion reaction of the incubation period, the number of twist body and body torsion score; And RT-PCR method is used to test the model in the rat spinal cord and the midbrain κ、μ receptors mRNA expression level of predominate.Results1 moxibustion Sanyinjiao(SP6), Guanyuan(RN4) on haemorrheological nature class dysmenorrhea body torsion reaction in ratsCompared with saline group, the writhing latency in model group was significantly shortened (P<0.01),the writhing number and writhing score in model group was increased significantly(P<0.01).Compared with the model group, the writhing number and writhing scorein Sanyinjiao(SP6) group decreased significantly(P<0.05); the writhing latency in Guanyuan(RN4) group wassignificantly longer(P<0.05), the writhing number and writhing score in decreased significantly(P<0.01). Compared with Sanyinjiao(SP6) group, the writhing number and writhing score in Guanyuan(RN4) group in decreased significantly(P<0.01).2 moxibustion Sanyinjiao(SP6), Guanyuan (RN4) on haemorrheological nature class dysmenorrhea κ、μ receptors predominate in the rat spinal cordCompared with saline group, group class dysmenorrhea haemorrheological nature card κ receptors predominate mRNA expression levels and statistically significant (P<0.05), spinal cord μ receptor mRNA expression level some edged higher, but no statistical difference (P>0.05); Two spinal cord κ receptors predominate mRNA expression level of moxibustion group were significantly increased (P<0.01), spinal cord μ, receptor mRNA expression level was increased (P<0.05). Compared with haemorrheological nature card type dysmenorrhea group, two spinal cord κ receptors predominate mRNA expression level of moxibustion group were significantly increased (P<0.01), spinal cord μ receptor mRNA expression level was increased (P<0.05).And Compared with Sanyinjiao(SP6) group, Guanyuan(RN4) group of spinal cord κ、μ receptors predominate differences had no statistical significance (P>0.05).3 moxibustion Sanyinjiao(SP6), Guanyuan (RN4) on haemorrheological nature class dysmenorrhea rat midbrain κ、μ receptors predominateCompared with saline group, group haemorrheological nature card type dysmenorrhea midbrain κ、μ receptor mRNA expression level was increased, but there was no statistically significant difference; Sanyinjiao(SP6) group and Guanyuan(RN4) group midbrain κ、μ receptor mRNA expression levels were significantly higher (P<0.01). Compared with haemorrheological nature card type dysmenorrhea group, Sanyinjiao(SP6) group and Guanyuan(RN4) group midbrain κ、μ receptor mRNA expression levels also were significantly higher (P<0.01). And compared with Sanyinjiao(SP6) group, Guanyuan(RN4) group midbrain preceptor mRNA expression level increased significantly (P<0.01).Conclusions1 General freezing method combined with recognized kind dysmenorrhea model method, successfully established the dysmenorrhea rat model with cold stagnation syndrome.2 All stduy acupoints (SP6, RN4) analgesic effect in dysmenorrheal model rats.3 Moxibustion the two points of Sanyinjiao(SP6) and Guanyuan(RN4) can significantly improve the model of rat spinal cord and the level of κ、μ receptor mRNA expression predominate in the midbrain.4 Speculated that moxibustion may be caused by promoting the spinal cord and the midbrain effective κ、μ receptor to generate and release, predominate the mRNA expression level increased, and activate the spinal cord level of two kinds of inhibition mechanism (presynaptic inhibition and prominent inhibition after) and the level of the midbrain descending inhibitory system, block nerve conduction of pain information, which help alleviate dysmenorrhea reaction of model rats.5 Different acupoints have different analgesic action. In this study Guanyuan(RN4) more clearly than the analgesic effect of Sanyinjiao (SP6), the possible reasons are:Guanyuan(RN4) with segmental innervation of the uterus more; Two different hole structure of local tissue, may cause the spinal cord and spinal cord, which is the central body and visceral afferent information gathering and synaptic connections between the different quality and quantity. Therefore, after moxibustion, formation of the afferent impulse as well as in the central excitability is different, Guanyuan(RN4) excitability is stronger; Guanyuan(RN4) acupoints in the abdomen, and uterine position close, the effects of the warm, radiation and drug moxibustion can directly infiltrate into the uterus, so as to better treatment effect.Above all, primary dysmenorrhea model with central nervous system κ、μ receptors predominate expression level of the disorder. Moxibustion treatment effect of PD on the part of the mechanism is by regulating the body of the spinal cord and the midbrain κ、μ receptors predominate to generate and release, and through the central inhibition of different level approaches to achieve the purpose of pain relief. And two different point have different adjustment effect of cellular palace, Compared with two acupoints and the adjusting effect of different cell palace,Guanyuan(RN4)point cave analgesic effect obviously.
