| Objectives:Dexamethasone injection and Lugua peptide injection on osteoblastproliferation and differentiationMethods: The rats were randomly divided into osteoblasts three groups, A groupadded1×10-7mol/L dexamethasone injection,8μg/ml Lugua peptide injection anddimethyl sulfoxide (DMSO, final concentration1g/L); B group is added1×10-7mol/L dexamethasone injection group A while adding an equal amount of DMSO; C withgroup A group only add the same amount of DMSO as a control group. After96h, byRT-PCR analysis of the expression of osteocalcin;After3d,5d,7d of ALP stainingand the activity of ALP detection; Contrast staining was observed after21days theexpression of calcium nodules.Results:1. Group A and group C mRNA expression of osteocalcin content werehigher than B group(P<0.01), and the A and control group C had no significantdifference (P>0.05).2. The ALP activity detection of group A and group C were higher thangroup B after3d,5d,7d (P <0.01).3.Group A and group C expression and calcium nodules were higher thanthose of group B (P<0.01), and the A and control group C had no significantdifference(P>0.05).Conclusion: Lugua peptide injection can fight under conditions of high concentrationsof dexamethasone inhibition of osteoblasts... |
