Objective:Evaluate the effects of the blood glucose of Type1diabetes mellitus SD rats that are under the route of administration of the dexamethasone subconjunctival injection and juxtaglomerular injection.Methods:Choose healthy SD rats, Male rats aged:2months, weighing220±20g, Were injected with streptozotocin in its abdominal cavity(Dissolved before using into the0.1mmol/L of pH=4.0citrate buffer,55mg/kg,1time), or injected with isodose citric acid buffer to establish Type1diabetes mellitus rat model SD.Select successful molded Type1diabetes mellitus SD rats (n=40) and normal SD rats (n40), and then divided them into group I Type1diabetes mellitus SD rats experimental group and group II normal SD rats control group. The Group I Type1diabetes SD rats experimental group were randomly divided into four groups and each group had10rats. Group I A belonged to subconjunctival injection of dexamethasone group; Group I a belonged to subconjunctival injection of saline group; Group I B belonged to injection of juxtaglomerular dexamethasone group; Group I b belonged to injection of juxtaglomerular saline group. The group II normal SD rats control group were divided into four groups and each group had10rats. Group II A belonged to subconjunctival injection of dexamethasone group; Group II a belonged to subconjunctival injection of saline group; Group II B belonged to injection of juxtaglomerular dexamethasone group; Group II b belonged to juxtaglomerular saline group.Rats were fasted without water for12hours, and then were anesthetized with2%sodium pentobarbital sterile liquid0.2ml/100g intraperitoneal injection. According to the group, they were injected with1-time0.1ml dexamethasone or with isodose saline under the bulbar conjunctiva or juxtaglomerular part. After injection, rats were fasted without water. Mining tail vein blood samples were collected. The use of blood glucose monitors (dipstick) assay3h,6h,12h,18h, and24h time point blood glucose levels in the eye during the local injection and after the injection.Results:Make use of intraperitoneal injection of streptozotocin, establish rat model of Type1diabetes mellitus SD, and test blood glucose after7days fasting, blood glucose level of successful model of Type1diabetes mellitus SD rats is (9.31±1.79) mmol/L and SD rats with normal blood glucose levels is (5.72±0.80) mmol/L after the injection of the isodose citrate buffer.For Group I Type1diabetes mellitus SD rats in the experimental group, the blood glucose level of injection of dexamethasone into the eyes in different ways(Subconjunctival and juxtaglomerular) was higher than saline group, Starting from6h after injection, the blood glucose differences between the injection of dexamethasone group and the injection of saline group were statistically significant (P <0.05). Within group comparison, it showed that dexamethasone group and the saline group had the highest blood glucose levels that are3h. the blood glucose level differences for the injection of dexamethasone group3h,12h,18h,24h and Saline group3h,6h,12h,18h,24h time point were statistically significant (P<0.05) comparing with before injection.Normal SD rats of Group Ⅱ in the control group, the blood glucose level of subconjunctival injection of dexamethasone was higher than that of the saline group, Starting from3h after injection, the blood glucose differences between the injection of dexamethasone group and the injection of saline group were statistically significant (P<0.05), Within group comparison, it showed that the blood glucose level differences for the injection of dexamethasone group3h,12h,18h,24h and Saline group3h,6h,12h,18h,24h time point were statistically significant (P<0.05) comparing with before injection. The blood glucose level of juxtaglomerular injection of dexamethasone was higher than that of saline group. Starting from3h after injection, the blood glucose differences for the injection of dexamethasone group and saline group were statistically significant (P<0.05), Within group comparison, it showed that the blood glucose level differences for the injection of dexamethasone group3h,6h,18h,24h and Saline group6h,12h,18h,24h time point were statistically significant (P<0.05) comparing with before injection.For the SD rats with Type1diabetes mellitus experimental group, whether the injection of dexamethasone group or saline group, the blood glucose level differences for the conjunctival injection and juxtaglomerular injection were not statistically significant(P>0.05).For normal SD rats control group, whether the injection of dexamethasone group or saline group, the blood glucose level differences for the conjunctival injection and juxtaglomerular injection were not statistically significant (P>0.05).Conclusions:1. In Type1diabetes mellitus and normal SD rats, Dexamethasone of subconjunctival injection and juxtaglomerular injection could lead to the rise of rats’ blood glucose The rise of blood glucose of SD rats with Type1diabetes mellitus was more obvious, which suggested that the injection of dexamethasone into the eyes would cause the rise of blood glucose.2. After saline was injected into ocular subconjunctival and juxtaglomerular eyes of the SD rats with Type1diabetes mellitus and normal SD rats. The blood glucose of SD rats with Type1diabetes mellitus elevated and the blood glucose of normal SD rats wouldn’t elevate, which suggested that local injection operation could cause the huge response in rats, and it made the rise of the blood glucose of SD rats with Type1diabetes mellitus.3. Dexamethasone of subconjunctival injection and juxtaglomerular injection led to the rise of Type1diabetes mellitus SD rats’blood glucose, but there were no differences in blood glucose values. It suggested that two kinds of methods had no differences for the effects of the production of rats’blood glucose.
