Malania oleifera is a peculiar single-species and single-category plant of our country. Its' seed has abound oil which is important raw material of oil in industry, so Malania oleifera has quite great value on exploitation and utilization. However, the currant natural narrow distribution and gradually rare resources of Malania oleifera restrict its' production and application. This paper studies comprehensively on the culture and repropagation of aseptic seedlings, the induction and subculture of calli and the cell suspension culture etc. of Malania oleifera. The preliminary experiments' results as follows:1. The explants are fresh seeds, which were peeled off seed coat. Through the surface disinfection on seed kernels, the suitable disinfection method is 10%NaClO10min+0.1%HgCl230min. The gaining rate of effective materials can reach over 90%. The seeds of Malania oleifera are hard to store and the germs are easy to loss water and wilt, which affect seriously the experiments' effects. So, the materials would be treated in 1 ~2 days after sampling. Moreover, the size of seeds also can affect the disinfection's effects.2. In the experiments of aseptic seedling culture and repropagation, the materials are unorganized germ and germ with endosperm, and the induction medium is blank MS. The results show that under our experiments' conditions, the unorganized germ cannot induct aseptic seedling, but the germ with endosperm can do, which induction rate is 85.3%. The optimum recipe of repropagation is MS+6-BA1.0mg/L+NAA0.2rng/L according to the results of subculture of aseptic seedlings. The reproduction multiple is 2~3.3. We have the embryonal axis, leaf and radicle etc. different positions of aseptic seedling inducted callus and subcultured. The results showed that the three materials all can induct callus, but the quantities of calli of embryonal axis are the most. The growing of these calli still did not improve after subcultered.Also, the three materials were differentiationed directly. All did not differentiation except for one hypocotyl which could induct bud on cambium.4. In the experiments of callus induction, we have the endosperms inoculated on six superior recipes, which basic medium is MS with 2.0-5.0 mg/L 2,4-D, 0.2- 0.5mg/L 6-BA and 0.2mg/L KT. The optimum induction recipe is MS+2,4-D5.0mg/L+6-BA0.5mg/L. In given consistency, 2,4-D with high consistency coorperated with 6-BA with low consistency can promote efficaciously the induction and propagation of calli. GA3 has no obvious function to the growth of calli. KT with suitable consistency can promote the growth of calli, so it is hoped that obtains better recipe through further experiments. The calli are light yellow, loose, fresh and lustrous. The six recipes were followed subculture. According the experiment results, the six recipes can be divided into three types: the first is earlier stage growth type, as MS+2,4-D 5.0mg/L+6-BA 0.5mg/L; the second is later stage growth type, as MS+2,4-D 2.0mg/L+6-BA 0.2mg/L+KT 0.2mg/L; the other recipes are medium type.5. The calli of endosperm were differentiationed, but they could not differentiation.6. The oil of callus on the six recipes was extracted and the oil rate was determined. The respective oil rates are 2.19%, 2.02%, 2.27%, 1.73%, 1.89% and 2.71%, which far less than the oil rate of seed kernels.7. In the experiment of cell suspension cultivation, the suitable shake speed is 100r/min, and the results of experiments of callus age of 30d are better than 45d. The optimum recipe of suspension cultivation is No.l. Also, the cell suspension cultivation of Malania oleifera has law as follows: when pH value ranges from 4.5 - 5.9, it is negative relativity between growth volume of suspended solids and pH value of slurry, namely pH value falls or rises along with growth volume reducing or increasing; Different recipes have different culture periods.8. The suspended solids were differentiationed. But we could not obtain optimum results.
