Objective To study the effects of operation by different anesthetic techniques on canine neuroendocrine and immune responses to stress , discuss mechanisms of immune depression at postoperation and provide therotical bases for alleviating stress reaction and immune depression during and after operation. Methods 24 crossbreed canine were alloted randomly into four groups: Xylazole general anesthesia (Group I ), Ketamine general anesthesia (Group II ), combination anesthesia of Xylazole and Ketamine (GroupIII), epidural anesthesia with Procaini hydrochloridi(Group IV) . White blood cell and its differential counts were carried out respectively before anesthesia induction , half an hour following incision , and at 4h , 1 st , 3rd , 7th and 15th after operation. At the same time , concentrations of globulin, cortisol and -EP, erythrocyte C3b receptor garland rates(RBC C3bRR), T lymphocyte blastogensis rates were measured respectively. Results White blood cell and neutrophil counts in every group increased at 4h after operation and at the 1st day after operation (P<0.05 ) , meanwhile lymphocytes decresed. Globulin content in every group decreased . RBC C3bRR in each group decreased and then increased except for Group I . T lymphocyte blastogenesis rates in each group decreased. Serous cortisol content in each group increased ,while cortisol content in Group I was higher than that in each other group (P<0.05) . Serous -EP content in each group increased and then decreased gradually, but -EP content in Group IV has a significant increased at half an hour following incision (P<0.05) .Conclusions Canine immune functions are suppressed by operation with different anesthetic techniques to different degree,but resume at 15th day after operation. There is an interacting and interadjusting mechanism among the CNS (central nerve system), the endocrine system and the immune system. Combination anesthesia of Xylazole and Ketamine can reduce stress response to anesthesia and operation, and immune depression caused by them.
