The most optimal combination of anaesthetic agent for dog we was determined by selecting present narcotics, sedatives, analgesics and other supporting drugs based on the latest medical anaesthetic theories and the physicochemical characteristics, mechanisms of action pharmacological actions of anesthetics, and according to the physiological characteristics of dogs after a series of scienti~c experiments. Pharmacodynamical action and practical application of the most optima] combination were also investigated in this study.Xylazoline, qiangtongning, peridol, ketarnirte were selected to be the agents used in this experiment; arid the preliminary composite ratio was also investigated according to the results of pre-experiment. The optimal probationary combination was determined through the four factorsthree level orthogonal tests and decisive orthogonal test with mice. Finally, the optima] combination and the most appropriate ratio were determined through anaesthetic effects test in dogs and security coefficient tests with mice. This optimal combination was temporarily named QMB combination.Acute toxicity tests were carried out by sequential method. The results showed that ED50, LD50of the QMB combination (small white mice i.p.) were 13.15 mg/kg, 67.676 mg/kg body weight and Al were 5.146, respectively. The results indicate that the Al is far more higher than usual demand (3). Therefore, QMB combination has a broad safety range of anesthesia.A dose of O.lSml桹.20m1/kg body weight of QMB combination was administered intramuscularly into clinically healthy mini-type dogs. The effects of QMB combination on respiratory, circulatory, cardiac and encephalic functions were investigated simultaneously undergoing operation experiments and monitored with a non-invasive way by multi-functional physiologic grapher 51-42, Datex respiratory monitor and circulatory monitor. The results showed that QMB combination produced balanced effects in sedative, analgesic, muscle relaxing, and had a slight influence on the physiological indexes of the dogs. Blood oxygen saturation was maintained above 90%, which indicates blood oxygen is plenty during the surgical anaesthetic period. These comprehensive effects are satisi~抜ng for the demands of ordinary, medium and large operations.Comparing QMB combination with 846 and iannaining?combination, the QMB combination is of superiority to 846 and iannaining?combination on the anaesthetic effects and less influences on respiratory, circulatory, encephalic functions.Postgraduate:Huanqi LiuMajor:Veterinary medicine Supervisor: Prof. Hongbin Wang...
