The frequent outbreak of animal diseases cost livestock producers millions of dollars in lost revenue, which also becomes a threat to human. One primary cause is lacking of the high efficacy, broad spectrum and low toxicity disinfectant. Based on the commonly used potassium peroxymonosulfate, organic acids, surfactant and an inorganic buffer system, a new compound disinfectant was developed in this study. It's important to prevent and control zoonotic pathogens and keep the sustainable development of our breeding industry. The study performed the quality control of sodium chloride and available chlorine in the compound peroxymonosulphate powder, based on which the stability of the compound peroxymonosulphate powder was investigated. It supported the scientific basis for the establishment of expiration time, suitable package and transport conditions. Finally, the pharmacological evaluation and toxicological evaluation of the compound peroxymonosulphate powder were studied to provide the theoretical evidence for the practical application. The results are as the followings:1 Quality control study of sodium chloride and available chlorine in the compound peroxymonosulphate powderTo develop a method for the determination of sodium chloride in the compound peroxymonosulphate powder, the test taked sodium chloride basic reagent as comparison, and a potentiometric-argentometric titration method was established to measure the contents of sodium chloride at normal temperature. The results showed that the linear range of the sodium chloride was 10.0 mg-55.0 mg with the correlation coefficient of 0.9997, the repeatability was good with RSD=1.11%, the average recovery with blank samples was 99.6% with RSD=0.88% and the average recovery was 99.1% with RSD=1.39%, which ensures its adequate accuracy. The method is simple and accurate. It could be adopted for the quality control of the contents of sodium chloride in the compound peroxymonosulphate powder.A N,N'-diethyl-p-phenylenediamine (DPD) method was established for the determination of available chlorine in the compound peroxymonosulphate powder. The sample was added into the mixture of DPD solution and phosphate buffer solution, then the red half-quinonoid compound formed by the reaction was spectrophotometrically determinated atλ= 325 nm. The results showed that the calibration curves were good linear between the absorbance and the concentrations of 0.05-2.0 mg/L with the correlation coefficient r = 0.9992, the repeatability was good with RSD≤2.65%, the average recovery with blank samples was 93.68%-105.55% with RSD≤1.29%, which ensures its adequate accuracy, and there was no significant differences in content of available chlorine between the DPD method and the iodimetric titration. The DPD method is simple and accurate. It could be adopted for the quality control of the contents of available chlorine in the compound peroxymonosulphate powder. To establish expiration time and suitable package and transport conditions for the compound peroxymonosulphate powder, the changes of sodium chloride and available chlorine were separately determined by potentiometric titration and iodometric method. The stability of the samples was tested by stress test, accelerating experiment and long term test. The results of stress test suggested that the compound peroxymonosulphate powder was sensitive to moisture. Double-layer aluminium foil was found to provide adequate protection for the product according to the accelerating experiment. After the first 12 months of the long term test, the compound peroxymonosulphate powder was demonstrated to be chemically and physically stable. The stability tests demonstrated that the compound peroxymonosulphate powder should be kept away from high light and may be stored for 2 years at room temperature.3 Observation germicidal efficacy of the compound peroxymonosulphate powder and its influencing factorsThe study was conducted to evaluate the germicidal efficacy of the compound peroxymonosulphate powder in vitro and to determine the effects of organic matter content, pH and temperature on its efficacy as a disinfectant, suspension quantitative germicidal test and animal test methods were used to carry out laboratory observation. The compound peroxymonosulphate powder demonstrated bactericidal activity against Escherichia coli, Klebsiella pnenmoniae, Staphylococcus aureus, Pasteurella multocida and Streptococcus suis at concentrations ranging from 0.03125% to 0.25%, the killing rate on the bacterial suspension exposed to 0.25% of the compound peroxymonosulphate powder solution for 5 min were all 100%, 1% CPP dissolved in ddH2O was not effective against B. cereus spores after 1h contact, but the sporicidal effect of the disinfectant was greater when dissolved in physiological saline solution (PS, 0.03mol/L NaCl) rather than in ddH2O; The minimal effective concentration of the disinfectant for killing E. coli and S. aureus in capacity test were 0.5%, The minimal effective concentration of the disinfectant for killing P. multocida and S. suis in capacity test were 0.1%; Furthermore, the disinfectant was highly effective at higher temperature and acidic pH. High fetal bovine serum concentration was found to compromise the bactericidal activity of the disinfectant to different levels in E. coli and S. aureus. The results demonstrated that the compound peroxymonosulphate powder had bactericidal activity against bacteria even at low concentration (1:3200). Increase in contact time, concentration of the solution and temperature or decrease in pH can improve the germicidal efficacy.4 The toxicological evaluation of the compound peroxymonosulphate powderTo evaluate the local irritation of the compound peroxymonosulphate powder, rabbit skin irritation test and rabbit acut eye irritation test were carried out. The results showed that a concentration of 1% CPP has no irritation to skin and eyes.In conclusion, the compound peroxymonosulphate powder has good stability, low toxicity, high germicidal efficency. the disinfectant is of great value to the environmental disinfection. 2 Stability of the compound peroxymonosulphate powder...
