| Gardenia is a traditional Chinese medicine, it has several important pharmacological effects, the content of gardenia yellow and geniposide is higher in Gardenia. Gardenia yellow is water-soluble and non-toxic, so it has been widely used in food field at home and abroad; Geniposide is the effective constituent of the gardenia for the cholagogic and anti-inflammatory effect. Its aglycon, genipin, not only has certain medical biological effects, but also can be used to manufacture gardenia blue, so at present geniposide is often applied to agriculture and medicine, etc.In this thesis, firstly, the gardenia yellow and geniposide mixture was extracted from water gardenia, then it was isolated and purified to obtain gardenia yellow and geniposide using macroporous resin; Afterwards, genipin was manufactured with geniposide as raw material and Aspergillus usamii as fermentation strains. Finally, the gardenia blue was prepared through the reaction of genipin and amino acids. The main experiment contents were as follows:1. Firstly, the determination methods of gardenia yellow pigment, geniposide, genipin, and gardenia blue pigment was established:the gardenia yellow pigment and gardenia blue pigment were determined using UV spectrophotometry; the geniposide and genipin were determined using high performance liquid chromatography.2. The optimal conditions for extracting the gardenia yellow pigment and geniposide from water gardenia fruit was established by single factor experiment and orthogonal design experiment:solid-liquid ratio1:8, ethanol concentration60%, extracting temperature60℃, extracting time1h, extracting times twice.3. In order to select the suitable macroporous resin to isolate the gardenia yellow and the geniposide, the ability of five different kinds of macroporous resin were investigated. According to the result, HPD100A was selected to adsorb the gardenia yellow and the geniposide, and then H103was used to purify the geniposide.4. The technology of separation and purification of gardenia yellow and geniposide with HPD100A was investigated, the process for future purification of the geniposide with H103was also studied. According to the optimized technology, the dark yellow gardenia yellow powder was obtained, and its OD value ratio was less than0.3and the colourity was350(E1cm1%,440nm), the high-quality yellowy geniposide which purity was more than90%was also obtained.5. The optimum process for preparing genipin with geniposide as raw material and Aspergillus usamii as fermentation strains was established by single factor experiment and orthogonal design experiment. The optimum process:0.2%geniposide as fermention medium, flask volume100mL/250mL,28℃,200rpm,66h. Following this project, the purity of the obtained genipin sample was more than99%.6. The process for preparing the gardenia blue through the reaction of genipin and amino acids was optimized. The optimum process:the ratio of sodium glutamate and genipin is5:1,80℃, stirring6h in water. Following this project, the black blue gardenia blue was obtained. |
