Glycerolipids involve in the distinguishing of cell membrane proteins and signal transduction. The production of signaling lipid is controlled by many lipid signaling enzymes. Phospholipase is one of the important lipid signaling enzymes. Activation of phospholipase modulates the output of signaling lipids to respond to the specific changed biotic or abiotic environment. Recently, six pupative phospholipase C genes were identified in Arabidopsis, which were named as nonspecific phospholipase C1 to C6 (NPC1 to NPC6) So far, only NPC4 was studied by Yuki Nakamura who pointed the expression of NPC4 is induced in the absence of Pi. In order to learn about the function of NPC4 from Arabidopssi in the developing process, we use the technology of RT-PCR at the RNA level and GUS staining at the protein level to study the expression pattern of Arabidopsis NPC4 . The main results are as follows:1. The expression pattern of NPC4 at the RNA level by using the analysis of RT-PCR By the analysis of RT-PCR in different developing period and different organs at Mrna level, we found that NPC4 gene highly expressed in senescing leaf and lowly expressed in root, stem, flower, siliques.In the early stage, NPC4 gene is not detected.This shows the expression of NPC4 gene is mainly in mature leaf and low in developing organs.2. The expression pattern of NPC4 at the protein level by using the analysis of GUS staining We clone 1379bp promoters of NPC4 upstream and fuse with GUS reporter gene.Then transform wild Arabidopsis and get the transgene plant.GUS staining found that the whole plant is not stained in the early stage.This shows the expression of NPC4 gene is low in developing stage. Meanwhile, in mature plants, it was stained.This shows the expression of NPC4 gene is mainly in mature organs, especially in leaf.3. The induced expressison of NPC4 gene The analysis of RT-PCR and GUS staining shows NPC4 gene is not induced by SA and MeJ, but induced by ABA.
