Taxonomic Identification Of Several Macro-Fungi Of Xinjiang And Study On Correlative Biological Characteristic

In this present work, five Micro-fungi in Xinjiang were studied, including morphology, anatomy, growth community and partial ribosomal DNA sequences. Internal transcribed spacers (ITS) or 28S large subunit domain 1 and 2 (LSU D1-D2) of five Micro-fungi were amplified and sequenced, comparing five fungi sequences with similar coreNucleotide data which have been submitted into international nucleotide pool of GenBank/EMBL/DDBJ/GSDB, BLAST (a kind of freeware provided at NCBI) program were applied. All results obtained were effectually proof to identify those strains to their genus or species level. In addition, phylogenetic trees based on CLUSTAL_W (version 1.6) analysis of all constructed tree species also were identified those strains. A new chinese record species of Emericellopsis pallida from Xinjiang was investigated, including morphology, cultivate and fermentation characteristics. The following were results.â…°Based on conventional morphology taxonomy, both XJURML4 and XJURML5 were identified as Morchella esculenta and Morchella crassipes, respectively. However, sequences of two strains in ITS and phylogenetic tree show them should be classified into Morchella esculenta(L.:Fr.) Pers.var.rotunda Pers.:Fr. Sequences of XJURML4 and XJURML5 are the same in ITS2, only 5 base pairs difference in ITS1.we hypothesis two strains produced small variance in similar growth community. RAPD bands profile of the two strains genomic DNA have also some difference. Data of ITS, phylogenetic trees and RAPD bands showed two strains were Morchella esculenta. GenBank access number are EU086775 and EU086776, respectively.â…±The morphology characteristics of XJURML11 were similar to Melanoleuca spp. Its sequences in 28S rRNA LSU D1-D2 have highly similarity with Melanoleuca melaleuca, similarity value was 97.79%. Combined morphology characteristics with molecular characteristics, XJURML11 was identified as Melanoleuca melaleuca, GenBank access number is EU621790.â…²Based on morphological characteristics, XJURML12 was primarily identified as Coprinus atramentarius (Bul1.)FT. Both 28S rRNA LSU D1-D2 and ITS of XJURML12 were amplified, but its double direction sequence didn't uncompatible. BLAST analysis of single wing sequence shown XJURML12 have highest homogenous with Coprinus rhizophoru (syn. Coprinopsis strossmayeri),whereas 28S rRNA LSU D1-D2 of XJURML12 have no matching data in international nucleotide pool. Because sequences of 28S rRNA LSU D1-D2 deprived of XJURML12, so we submitted integrated LSU D1-D2 sequence to GenBank and access number is EU621791.â…³Morphological characteristics of XJURML3 are similar to the reported species Emericellopsis pallida strain CBS 624.73 (AY632667), . homogeny was 98.33%. Based on microorganism nominate principle (up to 98%), XJURML3 was identified as Emericellopsis pallida and was a new variation. GenBank access number is EU045572, XJURML3 was deposited at China Center for Type Culture Collection and accession No.CCTCC AF 207025.â…´Cultivate and fermentation characteristics of XJURML3 were elucidated its biological Characteristic.Single factor method and response surface method were applied for optimal producing extracellular polysaccharide of XJURML3 (Emericellopsis pallida) among different hydrocarbons, nitrogenous and auxiliaries. The results obtained from single factor method were as preparative condition, and results of response surface method were used to systematic analyse extracellular polysaccharide. It was shown that the maximum yield of extracellular polysaccharide is 1.5g/l, with saccharose 59.5g/l straw juice2g/l and NH4Cl 2g/l. XJURML3 utilized straw juice very well, those optimum results illumined that response surface method prior to single factor method method, and response surface method rooted on single factor method...
