Having novel structures and diverse bioactivities,natural products from microorganisms have become an important source for new drug discovery.Many commonly used clinical drugs are originated from microbial secondary metabolites with bioactivities such as anti-tumor,anti-bacteria,antivirus and anti-oxidation.In this thesis,we have studied metabolites of a macrofungus(Panus rudis Fr.F01315)and a marine fungus(Penicillium brefeldianum WZW-F-69)using various of chromatographic techniques such as silica gel column chromatography,gel column chromatography,and high performance liquid chromatography and different analytical methods including NMR(Nuclear Magnetic Resonance)and MS(Mass Spectrometry),with the hope offinding compounds with novel structures and promising bioactivities.Macrofungus Panus rudis Fr.F01315 was cultured on PDA(Potato Dextrose Agar)medium.Twenty-nine compounds were isolated and elucidated,including fourteen new halogen containing compounds and their derivatives,eight ketone compounds having four new analogs,and other seven compounds having three new analogs.New compounds LHP-01-041-03,LHP-01-046-01,LHP-01-055-09 and LHP-02-028-01 showed board spectra of biological.activities against cancer cell lines including Kyse450,SKGT-4,A375,HL60,JEKO-1,MDA-MB-231.Furthermore,LHP-01-041-03 and LHP-01-046-01 exhibited excellent cytotoxicities against HL-60 and JEKO-1 cells.Ten compounds were isolated from marine fungus Penicillium brefeldianum WZW-F-69 cultured on rice medium.Majority of these compounds are indole derivatives including 4 indole diterpenes(F-69-02,F-69-03,F-69-04 and F-69-05),3 indole triterpenids(F-69-01,F-69-06 and F-69-07),and 2 macrolides(F-69-08,F-69-10)and 1 ketone derivative.In antiproliferation assay,compound F-69-10,a brefeldin A derivative,has IC50 values against U20S,SKGT-4,Kyse450 and MCF7 less than 4 ?mol/L.Furthermore,all of the new compounds were tested on other bioassays including anti-bacteria,anti-oxidation,anti tyrosinase and anti acetylcholinesterase assays.The results indicated that some of new compounds have antibacterial activity and anti-oxidation activity.
