Francois Langur (Trachypithecus francoisi) has been defined the First-Class National Protected Species in China and Endangered-Class in the world.It distributes restrictedly in the southwest of China and northern Vietnam. Guangxi province is one of the main distribution area of the Francois Langur in China,but their habitat fragmentation seriously destroy and the population size declines every quickly in Guangxi.Therefore understanding the morphologicaland molecular variation of different geographic group of Francois Langur is every important for us designing conservation strategies.we used faecal samples to investigate their Mitochondrial DNA, and 142 samples were came from the counties of Longzhou,Daxin,Long'an, Fushui in Guangxi and 3 were came from Guizhou.We used the sequence data of control rigion to investigate the current status of population genetic structure ofFrancois Langur in Guangxi.The results show :(1)Only 110 samples could used to analyse,the success ratio of PCR amplification was 75.86%.(2)The length of the D-loop of mtDNA genome from faecal samples was 395bp,43 sites were polymorphic,16 haplotypes were identified.(3)Among the Francois Langur population of four geographical groups,the Longzhou group have higher haplotype higher numbere of base differences, average number of nucleotide difference.(4)In short distance no obvious phylogenetic structure among haplotypes was found for samples of different origins,but in long distance the phylogenetic structure was found for samples of different area.(5)The network shows that the number of the variable sites between haplotypes is 1-32. Based on these results,we suggest that,in term of protecting gene diversity of Francois Langur in Guangxi we propose classifying the Longzhou group as the unit of high priority protection in the future.And enhance the Scotch cousin intercommunion of different groups,sometimes we must use ex-situ conservation.
