The research on the effect of calcium on phases of plant tissue culture was emphasized applied and was pilot studied for mutuality mechanism. It was included establishing optimal plant tissue culture systems for Biluochun Tea, Pyrus pyrifolia, Eriobotrya Japonica lindl, Photinia frasery, Lilium longiflorum, Ananas sativus, Sinningia speciose and Pelargonium citrosum Vanleenii. Among these systems, it is the first time to establish optimal plant tissue culture system for Biluochun Tea. Based on the eight plant tissue culture systems, the different concentrations of Ca2+ were supplemented into the media to test their effects on proliferation and rooting. Along with the rise concentration of Ca2+ (0~12 mmol-L-1), it was found that Pelargonium citrosum Vanleenii proliferation period can be prolonged . In proper concentration of Ca2+(3~12 mmol·L-1), premature senility was restrained, SOD was improved and protected, and MDA was reduced for maintaining membrane stability. The Ca2+ levels in the test-tube plantlets were measured, and the results showed that Ca2+ presented positive correlation with those in the media. That the suitable concentration of Ca2+ (3~4.5 mmol·L-1) were supplemented into the media can improve rooting in early phase for some kinds of difficult rooting plantlets upon with the base concentration of Ca2+ (1.5 mmol·L-1).
