Tissue Culture Of Three Plants And The Changes Of Enzyme Activity During Rooting

In the study, Acer platanoides'Crimson King',Euphorbia phlcherrima and Hemerocallis hybrida were used as explants for tissue culture and the efficient plant regeneration system were established.At the same time, the effects of different plant growth regulators on growing,shoot propagation and rooting of Euphorbia phlcherrima were studied.Finally,we studied the changes of enzyme activity and peroxidase isozyme spectrum during rooting.The results showed that the most suitable mediun for shoot propagation of Acer platanoides'Crimson King'was MS+BA1.5+ZT0.1+NAA0.1+GA0.5+CH300 and the shoot propagation rate was 4.35;The best mediun for rooting was MS+NAA0.1+ IBA 0.5+PG40 and the rooting rate was 72.4%. The best medium for shoot propagation of Euphorbia phlcherrima was MS+BA1.0+IBA0.1 and the shoot propagation rate was 7.87; The promoting root medium was 1/2MS+IBA1.0 and the rooting rate was 82.6%; The ideal medium for differentiation of adventitious buds of Hemerocallis hybrida was MS+BA1.0+KT1.0+IBA0.5 and the differentiation rate was 93.3%;The medium of 1/2MS+IBA0.5 was proper for rooting of tube seedlings and the rooting rate was 100%.The results showed that DPC of lower concentration had effect on enhancing multiplication of the shoot of Euphorbia pulcherrima.Adding 10mg/L DPC can promote the growth of seeding and the tube plantlets became stronger.Tube plantlets had stronger and lower stem under the treatment of 30mg/L DPC.Compound Vitamine B at 0.5~1.0mg/L were fit for increasing the height of tube plants. Compound Vitamine B at 0.2~0.5mg/L could accelerate root differentiation and increase the amount of the root.The results showed that the change trends of IAAO and POD in Euphorbia phlcherrima and Hemerocallis hybrida were similar.While the change trends of IAAO and POD in Acer platanoides'Crimson King'were different.It also conclued that when IAAO and POD activities were at the lower position of the activity curves benefit rooting.The original activities of the enzyme in the plants maybe an influence factor,which dedermine the rooting ability.The peroxidase isozyme spectrum has changed during rooting stages.
