Arabidopsis thaliana has good characteristics of small genome, simple structure, smallplant, short life period, high propagating rate, self-pollinating etc. And the whole genomesequencing has been completed in December 2000.We have established methods forscreening for mutants of Arabidopsis with enhanced resistance to Oxalic Acid. The mainresults were as follows:1.The Arabidopsis seeds were cultured in medium containing with oxalic acid forscreening mutants with enhanced resistance to oxalic acid. The proper screeningconcentration of oxalic acid was 1.2mmol/L.2.Five A. thaliana mutants with enhanced resistance to oxalic acid were obtained afterscreening two times in medium with 1.2mmol/L oxalic acid. The T-DNA flankingsequences of the 5 mutants were amplified by TAIL-PCR and blasted with Arabidopsisdatabase at the TAIR website or the MIPS website. From the blast results, the T-DNAinsertion sites were found. Among them, T-DNA of four mutants (D630,282,D154,D74) inserted in the same locus: one intron of At5g10450.T-DNA of the fifth mutant inserted inthe region locus between At2g39720 and At2g39730 genes.3.The loss-of-function mutant of At5g10450 gene was gotten from ARBC and it wasnot resistant to oxalic acid.4.The over-expression vectors of At5g10440 gene and At5g10460 gene wereconstruted and transferred into wild type plants of Arabidopsis. The transgenic plants withbasta hcrbicide resistance were scrcened out.5.The over-expressing vector of oxalate oxidase gene was construted and transferredinto wild type plants of Arabidopsis.. The transgenic plants with basta herbicide resistancewere also obtained.6.The oxalic acid resistance mutants and wild type plants were inoculated withpathogen of Sclerotinia sclerotiorum. The results showed that the mutants enhancedresistance to S. sclerotiorum.In summary, there is a relativity between the resistance to oxalic acid and S.sclerotiorum in plant.
