Analyse genetic polymorphism in some STR loci and genetic structure in Hui and Han population in Ningxia.The genetic structure of a population is the allele frequency of all loci. Genetic structure difference is virtually difference of gene frenquency, which leads to the diversity of human genome. Research on human gonome diversity is important for researches to find differences between individuals as well as populations. And it also very helps a lot in pathological gene discovery and genetic disease researches.The genetic polymorphisms of 9 short tandem repeat (STR) loci of D5S818, D7S820, D8S1179, D11S1984, D14S306, D14S617, D16S539, D17S1290 and D19S433 in unrelated individuals in Hui and Han population in Ningxia area are analyzed by PCR amplification, denaturing polyacrylamide gel electrophoresis (PAGE) and silver staining; and using the gene frequency data of 4 STR loci of D5S818, D7S820, D8S1179 and D16S539, genetic distance are calculated between Ningxia Hui and Han population, between Ningxia Hui and Han in other districts and between Ningxia hui and minority populations in other districts in order to analyse the genetic structure by cluster analysis and principal components analysis.The result shows that 9 STR loci have better polymorphism in Hui and Han in the Ningxia area except D7S820. The heterozgosity of the 9 STR loci are 0.7074-0.8437, the polymorphism information content are 0.6683-0.8244, the discrimination power are 0.8680-0.9660, the probabilities paternity exclusion are 0.4955-0.7079 in Ningxia Hui population; the heterozgosity of the 9 STR loci are 0.7726-0.8444, the polymorphism information content are 0.7388-0.8265, the discrimination power are 0.9066-0.9484, the probabilities paternity exclusion are 0.5880-0.7123 in Ningxia Han populatioaThe principal component analysis and cluster analysis in 4 STR loci shows that there are some genetic structure differences between Ningxia Hui and Han population, between Ningxia Hui and Han in other districts and between Ningxia hui and minority populations in other districts.The genetic polymorphism analysis indicates that the 9 STR loci we chose have better polymorphism in Hui and Han populations in Ningxia district except D7S820. At the same time, the 8 loci have their own distribution characters, and thus could serve as useful markers in population genetics research, individual identification and paternity test in forensic medicine. The analysis also indicates that D7S820 is not a good genetic marker for individual identification and paternity test in Ningxia Hui populationThe results of genetic structure analysis using 4 STR loci indicate that Ningxia Hui has its own genetic structure character and there are some differences between Ningxia Hui and Han as well as other minorities. But there also exists gene flow and fusion between Ningxia Hui and central China Han as well as Ningxia Haa On the contray, there are little differences between Ningxia Han and Han in other districts in China especially in North China.
