Solid Culture Conditions Of Azadirachta Indica A. Juss And Effect On The Content Of Azadirachtin

The neem tree {Azadirachta indica A.Juss) is one of the most promising carriers of botanical insecticides. Substance from neem has the nature of high efficient insecticide and fungicide. Azadirachtin is its main active substance. Neem can not distribute extensively in China, so we need to find a new way to exploit the neem tree. Standing on this point, this paper maily studied on tissue and cell culture of neem for Azadirachtin. The results follow as below.1. Using leaf and twig as explants, we get high inducing frequency of calli, but seeds that hasdeposited for half a year have a very low inducing frequency. Considering the inducing frequency and growth of neem calli, the MS medium is the best choose.2. B5+NAA(1.0)+ IBA(1.0)+ABA(1.0) is the best combination of ingredient in solid culture. All thecalli from different explants once has a high content of Azadirachtin, but different explants-source needs different culture condition, calli cultured on MS medium showed a wide range of Azadirachtin content, but B5 medium, the range is much narrower. ABA is the key factor in neem's in vitro culture. pH6.0, 14h light/1 Oh dark are the appropriate option for neem's in vitro culture.
