This paper studied the effects of types of medium, conditions of culture, different kinds of carbon and nitrogen sources and plant growth hormone in medium component on Agave sisalana tissue and cell culture for inducement of Agavain -SH. Mutagenic radiation with Co60-γ ray was adopted and cell suspension culture of callus were primary studied. The main results are as follows:1. Callus inducing and culturing of Agave sisalanaMS LS N6 B5 mediums were adopted to study callus inducing and growing of Agave sisalana. The results shown that the callus first produced in the N6 medium. In LS and MS mediums callus inducing rates were high and growth were good. B5 mediums was not fit for the callus inducing and growing . The callus grown well under the temperature of (25±2)℃.Under 12h weak light and 12h strong light illuminance the callus produced early and grown well. 24h illuminance and 24h dark were not suitable for the callus producing and growing.2. Callus Culture of Agave sisalana for the inducement of Agavain-SH. MS LS N6 B5 mediums were adopted to culture the callus for inducingAgavain—SH. It was shown that no matter the cell solid culture or the cell suspension culture , the callus cultured in 4 kinds of medium could all produce certain amounts of Agavain—SH. But the activities in MS and LS medium were higher. Two cell lines were selected with the activities 9.38 % and 12.51 % higher than the explant. Agavain—SH induced from the callus cultured in four kinds of mediums could secrete 20.01 57.23% of them into mediums.When the callus cultured under different temperature , i. e. (32±2)℃, (25±2) ℃ and (18±2)℃, the special activity of Agavain — SH improved as the temperature drop. High activity of Agavain—SH could be induced under 12h...
