Ma.ndarmfish(Siniperca chuatsi) culture has played an important role in Chinese aquaculture,while at the same time,many problems are still existing,and selection of good breed is the most important one . Qiupuriver Mandarinfish is a local breed under selecting and a famous brand of Chizhou city,Anhui province,which has a big reproductive population and culturing on a large scale;it also possesses of sound features such as high growth rate and strong resistance .But none study has been carried on its genetic basis before. Study on the genetic variation of Qiupuriver Mandarinfish,Changjiang Mandarinfish and Wanfuhu Mandarinfish with isozyme and RAPD Markers has important content,at the same time,it will bring new material to selection and application of the Qiupuriver Mandarinfish.First,three isozymic systems(Lactate dehydrogenase(LDH),Esteraes(Est),Malate dehydrogenase(MDH)) extracted from threepopulations of Mandarinfishes were detected by discontinuous vertical plate polyacrylamide gel electrophoresis(PAGE). Several loci were tested but none polymorphic locus was detected in Qiupu river population. The results showed that:LDH can be used as biochemical markers to identify these three populations of Mandarinfishes.Second,genomic DNA polymorphisms of three Mandarinfishes populations(each bulked DNA pool composed by 10 individuals) were tested by Rondomamplified polymorphic DNA(RAPD) using Bulked Segregant Analysis(BSA).12primers selected from Operon KitZ yielded reproducible and polymorphic DNAfragments ranging from 125bp to 3kb.These primers generated 60 scorable marker bands of which 15(25%) were polymorphic in Qiupu river population;59 bands in Changjiang population,15(25.4%) were polymorphic;and 58 bands in Wanfuhu Mandarinfishes,19(32.8%) were polymorphic. Bands produced by a single primer varied from 1 to 9.The stastical analysis of the experimental results indicated that:l.The intra-population genetic similarity index of Qiupu river Mandarinfishes,Changjiang Mandarinfishes and Wanfuhu Mandarinfishes were 0.9063,0.8839,0.8931;and the inter-population similarity index were 0.8590,0.8178,0.7956,respectively. Qiupu river Mandarinfishes have higher intra-population similarity index value than the other two populations,which showed that there were less genetic variations in Qiupu river population,as well as differentiations have been generated between Qiupu river Mandarinfishes and Changjiang Mandarinfishes.2. The genetic distance between Qiupu river population and Changjiang population is the cloest,which is 0.0411,and genetic distance between Qiupu river population and Wanfuhu population, between Changjiang population and Wanfuhu population is 0.1766,0.1230 respectively. It indicated that Qiupu river Mandarinfishes have a more closer relationship with Changjiang Mandarinfishes than Wanfuhu ones,which corresponding to the clustering analysis and their geographical distribution.3. With the primer OPZ05,population-specific fragments were produced among three populations,and could be used as molecular genetic markers to identify them.
