The peroxidase isozyme(POD) and esterase isozyme (EST) of ten species of wild Poa in Gansu province were studied by means of polyarcylamide ? electrophoresis. It is successful to use improved SDS method to extract Poa general DNA,the method is suitable for Pea moleculebiology study. The optimum RAPD reaction system in Poe plant was settled. Thirty decamer oligonucleotide random primers from Operon Technologies were used for polymorphic selection,and 18 primers were selected for RAPD analyses. Data from the results of isozyme zymograme scaning and RAPD information were used for analyses. By Statistical program,genetic distance were generated and dendrograni was constructed using UPGMA.It is concluded as follows; (1) Extensive genetic diversity were present among the species. POD present zymobands at eight loci at most and four loci at least;EST present zymobands at eight loci at most and four loci at least. Thirty decamer oligonucleotide random primers were used for polymorphic selection, 28(93.33%) produced polymorphic RPLPD products. A total of 250 bands with 205(82%) polymorphic amplified were selected for RAPD analyses. (2) The cluster results obtained by RAPD analyses were basically comparable to morphology classification;and by POD isozyme analyses were similar to morphology classification;but those obtained by esterase isozyme were different from morphology classification. These revealed that RAPD analyses and POD analyses are the racid effective methods on Poa's classifi鏰tion,EST analyses is limited on Poa's classification.
