RAPD analysis was applied to study genetic diversity of Amphioxus(Branchiostoma belcheri tsingtaoense) in the test. Total twenty-two samples, which are eleven males and eleven females were collected for use.Of 40 random oligonucleotide primers (20 OPC & 20 OPA respectively) for ampliation of Amphioxus genomic DNA, 17 could produce distinctive and reproducible fingerprints varying in length from 200 to 2200 bp. One hundred and twenty-seven sites were detected, of which 60 of them (47.24%) were polymorphic. The average genetic distance in Amphioxus was 0.1344, and the range of genetic similarity indexes was from 0.78 to 0.96, the average was 0.8656. Each primer used for the detection could produce 4-11 molecular markers. The value of average heterozygosity was 0.1165. The result illustrated that there were low polymorphism between Amphioxus. Lacking of genetic diversity challenged the existence of Amphioxus on the decline in resources.Obvious difference bands can be detected between male and female from the electrophoresis. It is recorded OPC-07250 & OPC-12250. It's estimated there're some relations with hetero-sex-chromosome which male Amphioxus have. This coincides with the XY sex determination. This experiment provide the gists on the molecular level for studying and protection of Amphioxus.
