Richardson (Pseudosciaena crocea) was one of the "four biggest marine products". In the middle of 1970s, the resource of Richardson was deadly destroyed because of ceaseless catching. The technology of manual breeding Richardson promoted the industry of breeding Richardson in cage in 1990s. But at the same time, the resource of caged Richardson was degenerated badly in the case of inbreeding. In order to know about the resource of Richardson late years, we studied the genetic diversity of two caged stocks from Zhejiang and Fujian province, and two geography populations using RAPD in this article.Twelve primers produced eighty-seven loci, ranging in size from 280 to 1500bp, and the polymorphic loci was 36, the percentage of polymorphic loci was 41.38%. Nei's gene diversity, Shannon information index and the percentage of polymorphic were all indicated that the genetic diversity of population min-yue was higher then the population dai-qu. The order of the genetic diversity for the four caged stocks was: Pingtan>Luoyuanwan>Zhoushan>Ningbo. Genetic distance between two geography populations was 0.0462, and genetic identity was 0.9458. Gst value was 0.1435. Nm value was 2.9835. Analysis of molecular variance with distances among individuals showed 14.58% variation (P<0.001) between geography populations, and variation mostly from individuals within populations. The variation in caged stocks was high (82.38%).Richardson had a low genetic diversity. The caged populations went through a bottleneck during their history, so they had a closer relationship than the two geography populations. Nm value between two geography populations proved gene flow among the populations was or is exist. The low Gst value also proved genetic variation mostly within the stocks. Gst value (0.1435) between geography populations showed Richardson is a structured population in the history.
