Artemia sinica lives in the high-salt salt lakes and salterns which exists in Qinghai, Xinjiang, Tibet , Shanxi and Inner Mongolia. It belongs to Phylum Arthropoda , Class Crustacea , Subclass Branchiopoda , Order Anostaca , Family Artemiidae and Genus Artemia. Osmoregulation is essential for Artemia sinica survival after hypertonic stress. In the different development stages ,osmoregulation is accomplished by different tissues. When Artemia is hatched into nauplius,it is accomplished by salt gland and thoracic epipods show their affects in the sub-adult and adult stages. Signaling pathways of Ste20 kinase have been identified in yeast,but it is uncleanly in low lifes and animals.Our research makes a contribute for signaling pathways of Ste20 in hypertonic stress.After blasting a mRNA sequence of Ste20 in an EST library of Franciscana,they have the same conserved domain which called ATP-binding site. RT-PCR primer sequences of Ste20 were designed according to a part of identified sequence of Ste20,which were used for semi-quantitative RT-PCR of the same samples in different salinity.We find the expression quantity of Ste20 increases by degrees in the aggrandizement of salinity. By the immunohistochemical method to determine the expression site of Ste20 kinase in different developmental stages of Artemia sinicaThe results shows that evenly expressed in resting eggs in 0h ,5h and 10h; the expression for Artemia sinica in 15h ,20h and 40h centralizes mainly in nauplius cephalothorax when there is a little in abdomen; the expression for Artemia sinica in 3d centralizes mainly in cephalothorax, abdomen and enteric canal; the expression for Artemia sinica in 5d and 7d centralizes mainly in enteric canal and appendage.We use real-time PCR method to analysis the expression of Ste20 kinase in different development stages. The results shows that it is different of the expression of Ste20 kinase in different development stages. The fluctuation of the expression of Ste20 is not sharp in all development stages except for 20h. Ste20 kinases play a important role not only in systemic osmotic homeostasis,but also function in cell-cycle control, development and cell growth.
