Hypoxia up-regulated 1(hyou1)is a stress-related protein and closely related to diapause in the development of Artemia.In the present paper,the full-length 2952-bp cDNA sequence of the hyou1 homolog of Artemia sinica(As-hyou1)was isolated by RACE technology for the first time.This gene encodes 886 amino-acid proteins,belong to NBD sugar kinase HSP70-actin family members.The expression pattern and location of the As-hyou1 gene was investigated in different stages of the embryonic development by means of quantitative real-time qPCR and in situ hybridization(ISH)assay.As-hyou1 was distributed throughout the whole body in cells of different embryonic development of A.sinica as shown by ISH.This is followed by a dramatic downregulation after diapause and is newly upregulated from the larval nauplius stage.Furthermore,As-hyou1 transcripts are downregulation under conditions of high salinity and low temperature.These findings suggest that As-hyou1 is stress-related and may act as a influence factor in embryonic development.In situ hybridization analysis showed that As-hyou1 is expressed in a widely dispersed pattern before incubation and is mainly concentrated on the cephalothorax,abdomen,body surface and appendage,after the nauplius stage.Our results suggest that As-hyou1 is integral to the process of diapause and diapause termination in A.sinica. |
