Artemia sinica belongs to Phylum Arthropoda , Class Crustacea , Subclass Branchiopoda , Order Anostaca , Family Artemiidae and Genus Artemia. It lives in the high-salt salt lakes and salterns which exist in Shanxi, Tibet, Xinjiang, Qinghai and Inner Mongolia. Osmoregulation plays an important part in keeping its life. In the early embryo development stages of Artemia sinica, osmoregulation is accomplished by salt gland or/and intestines. This article makes a research for the expression location of Artemia sinica which are cultivated respectively in 0h, 5h, 10h, 15h, 20h, 40h and 3d by conventional immunohistochemistry and unitary immunohistochemistry technology, and makes a research for the expression quantity of Artemia sinica in different conditions by Western-blot. In the world, many scholars have committed themselves to the research of NFAT5, and the resarch for mammal is the most clear. In low lifes, the resarch of NFAT5 for fruit fly is the most clear. The quantity of researchs for genes which make a correlation with osmoregulation is very much, but there is no resarch for NFAT5 of Artemia sinica not in China but also in foreign country. This experiment makes a research for NFAT5 of Artemia sinica for the first time, and opens out the relation ship between NFAT5 and Artemia sinica antisaline. It makes a contribute for Developmental biology and Animal medicine both in theory and actual application.This research finds two results as follows: 1.Expression for Artemia sinica in 0h and 5h centralize mainly in gastrulae endoblast ; expression for 10hs'centralizes mainly in membrance inner nauplius endoblast and a little in membrance inner nauplius top. Expression for Artemia sinica in 15h distributes in head and salterns , and centralizes mainly in head ; expression for 20hs'centralizes mainly in head and salterns;expression for 40hs'centralizes mainly in head, some in salterns; expression of NFAT5 for Artemia sinica in 3d centralizes mainly in head, a little in salterns. As a whole, expression of NFAT5 for Artemia sinica's grub and imago centralizes primarily in head and salterns, and centralizes most mainly in head. 2. Finding that in the condition for the same temperature, same salinity and same illumination intensity, the change for NFAT5 expression quantity of Artemia sinica in 0h,5h,10h, 15h,20h,40h and 3d is primarily unconspicuous, namely, the fluctuation is not sharp. Contrarily, in the condition for the same temperature, same illumination intensity, same period (Artemia sinica in 3d) but different salinity , we can find that as the aggrandizement of salinity, NFAT5 expression quantity increases by degrees. This shows that NFAT5 expression is intensively influenced by salinity change, and plays an important role in regulation of salinity change.
