| Background:Rat is a normal laboratory animal among mammalia laboratory animals. because of its less body model and its genetic character and longevity are similarity to experiment condition, It was called exact biology tool which was used to biomedicine research. More than 50 rat varieties were breeded by researchers on the basis of about 100 mutation genes were discovered. These animal models which were developed by people were useful in the research of human disease. Curly mutation was a mutation which hair and vibrissae were curly in rats. There were not reported by research in our country, but some curly mutation types were discoverd in outside of our country. Most curly mutations were caused of the mutation of EGFR gene or TGFagene. These mutant genes affected hair follicle growth in curly rats. Other phenomenon such as the heteroplasia of sebaceous gland, galactophore, heart and skelecton et al. There were more significance in these mutations to the research of upgrowth, tissue differentiation, growth regulation and the disease of heart or other apparatus.Yuyi Curly hair rat (YCHR) was discovered from SD rat species by laboratory animal center of Henan province in 2006. There were three types, such as normal hair, curly hair, and sparse and curly hair, in YCHR's offspring. These rats could be distinguished easily from other rats in 2 or 3 days after birth because of its curly vibrissae. In our study, There were some difference in phenotype and genetic character between YCHR and SD rat. We have breeded 11 generation of YCHR, but have not understand molecule mechanism in these curly mutations. In this research, we use some methods of genetics, pathology, immunology and molecular biology et al, to discuss the skin character and gene mutation mechanism in YCHR.Methods:we used the technique of pathology and immunohistochemistry to research the skin of YCHR and normal SD rat. Moreover, we analysed the cDNA sequence of EGFR gene. We also study on homology comparison betweent YCHR and other mammals at nucleotide on the basis of EGFR gene.Results:Compared with that of SD rats, backskin from Yuyi curly rats revealed a dramatic disorientation of hair follicles, and EGFR expression with Structural Changes in the YCHR Skin as Revealed by immunohistochemistry. Moreover, we achieved the cDNA sequence of EGFR gene and discovered 3 mutation sites in this nucleotide sequence.Conclusion:There were curly hair follicle in the skin of YCHR. We think this change may be the efficient amse of curly hair. we discovered some mutation sites in EGFR gene.The mutation of EGFR gene may be one of the curly hair reasons in YCHR. |
