Based On The Theory Of "preventive Treatment Of Disease" Of Traditional Chinese Medicine To Explore The Regulation Of "Winter Disease Being Cured In Summer" On The People And Intestinal Flora Of Specific Endowment Constitution

Objective: 1.Through systematic review and meta-analysis of the literature on the treatment of the familiar disease of specific endowment constitution with " winter disease being cured in summer ",this paper provides a reference for the clinical application of " winter disease being cured in summer " in the treatment of the familiar disease of specific endowment constitution.2.By analyzing the specific endowment constitution's tongue,face,pulse diagnosis and intestinal flora,exploring the physical characteristics and intestinal flora of the specific endowment constitution,in order to explaining the specificity of specific endowment constitution in terms of intestinal flora.3.Through the research on the regulation and control of Dog Days plaster in specific endowment constitution and intestinal flora,the theory of " winter disease being cured in summer " of traditional Chinese medicine is strongly supported by experiments,and the clinical application of Dog Days plaster is expanded and guided,so that Dog Days plaster can be used scientifically and benefit mankind.Method: 1.Systematic review and meta-analysis of literature on "treating winter diseases in summer" for the familiar disease of specific endowment constitution(1)Information retrieval : Literatures were retrieved from CNKI,WANFANG DATA,VIP and Pubmed.Chinese search is based on the combination of subject words and keywords,and foreign language search is based on the combination of Mc Gregor's words and subject words.(2)Literature inclusion and quality evaluation: Eliminating repetitive literature,and then according to the document title and abstract,eliminate the literature which does not meet the inclusion criteria.Finally,read the full text of the remaining literatures,and select the qualified literatures according to the inclusion and exclusion criteria.According to the improved Jadad scoring,collecting statistics and evaluating the quality of the selected literature.(3)Literature data collection and statistical analysis: Systematic review and meta-analysis were performed using Review Manager(Rev Man 5.4)software.Since the outcome indicators were all dichotomous variables(counts),the effect size was determined by relative Risk Radio(RR),and point estimates and 95%CI were given for each effect size.The heterogeneity of the included studies was analyzed by chi-square test(test level was ?=0.1)and evaluated in combination with I2 statistics.If the heterogeneity test result I2 was less than 50%,the fixed effects model was used for meta-analysis.If the heterogeneity test result I2 is greater than or equal to 50%,it indicates that there is statistical heterogeneity among the results of various studies,and further analysis of the source of heterogeneity is required.After excluding significant clinical and methodological heterogeneity,a random-effects model was used for metaanalysis.Sensitivity analysis was to perform meta-analysis again after removing a single study,and evaluate the difference between the results after elimination and the combined results of the original results.Publication bias was intuitively judged by drawing funnel diagram and quantitatively evaluated by Egger's test linear regression method using Stata 12.0 software,and the test level was ?=0.05.2.Based on the theory of "preventive treatment of disease" in traditional Chinese medicine,discussing the regulation effect of Dog Days plaster on intrinsic constitution.(1)Subject inclusion: According to the inclusion criteria,subjects with specific endowment constitution were included,and the "random number table" was used to randomly divide the subjects into two groups.(2)Intervention method: Subjects were applied application on the first day of each "dog days of summer"(mid-July,late July to early August and midAugust)for 6 times in 2 years,and each time for 4 hours.The experimental group used Dog Days plaster,the control group used comfort application,and both subjects and researchers were blinded during the trial.(3)Collection of samples and indicators: The Constitution determine, surface conditions,tongue conditions,pulse conditions as well as the feces were collected within 1 week before the intervention,6 months after the completion of the first intervention and 6 months after the completion of the second intervention.3.Based on the traditional Chinese medicine theory of " preventive treatment of disease ",discussing the regulation of Dog Days plaster on intestinal microecology of the people in specific endowment constitution in Xindu district.(1)Extraction and identification of gut microbiota: After gut microbiota was extracted with the kit,16 S r DNA sequence sequencing was used to identify the species of bacteria in the samples.(2)Bioinformatics analysis and statistical analysis: The diversity of intestinal flora was analyzed by OTU clustering analysis,species classification annotation,Alpha diversity analysis,Beta diversity analysis and significant difference analysis of community structure.SPSS 26.00 was used for data statistical analysis.Result: 1.In the meta-analysis of allergic rhinitis,the treatment effect of " winter disease being cured in summer " group was better than that of the control group;The therapeutic effect of Dog Days plaster combined with Chinese medicine or Chinese patent medicine was better than that of Chinese medicine or Chinese patent medicine alone.The therapeutic effect of Dog Days plaster combined with western medicine was better than that of western medicine alone.After six months and one year of intervention,the therapeutic effect of Dog Days plaster combined with Chinese medicine or Chinese patent medicine was better than that of Chinese medicine or Chinese patent medicine intervention alone.The therapeutic effect of Dog Days plaster intervention alone is better than that of acupuncture and moxibustion alone,but it cannot be explained that there is a difference between the therapeutic effect of Dog Days plaster combined with acupuncture and moxibustion and that of acupuncture and moxibustion alone.In the meta-analysis of cough variant asthma,the treatment effect of "summer treatment for winter disease" group was significantly better than that of the control group.The therapeutic effect of Dog Days plaster combined with Chinese medicine or Chinese patent medicine was better than that of Chinese medicine alone.The curative effect of Dog Days plaster combined with conventional treatment was better than conventional treatment alone.The therapeutic effect of Dog Days plaster combined with antihistamine was better than that of antihistamine alone.In the meta-analysis of classic asthma,the treatment effect of "summer treatment for winter disease" group was significantly better than that of the control group.The therapeutic effect of Dog Days plaster combined with conventional Western medicine treatment was better than that of conventional Western medicine treatment.2.Most of the people in specific endowment constitution present normal color and less gloss in face,red lips,dark red and white light tongue,white and thin furred tongue,the shape of the tongue is moderated in size.The local feature of face color and tongue color,as well as the feature of furred tongue and shape of tongue are no specific.Most of the left pulse presents Xihua Mai and Ping Mai,while most of the right pulse presents Xihua Mai and Hua Mai.3.After the initial intervention of the experimental group,the red and light white complexion decreased,while the normal complexion increased(*p<0.05).In left pulse,the proportion of Chi Mai,Hua Mai,Xihua Mai,Xixian Mai,Chenhuan Mai,Shuo Mai,Huanxian Mai,and Chenxixian Mai has decreased,the proportion of Hua Mai,Chenxihua Mai,Ru Mai,Ping Mai and Fushuo Mai has increased,the proportion of Xian Mai and Xianshuo Mai has unchanged(*p<0.05).The score of specific endowment has significantly decreased(** p<0.01).After the second intervention of the experimental group,pale,red and light white complexion decreased,white complexion increased,and normal complexion increased significantly(**p<0.01).In left pulse,the proportion of Chi Mai,Xihua Mai,Xixian Mai,Chenhuan Mai,Shuo Mai,Xian Mai and Huanxian Mai has decreased,the proportion of Hua Mai,Ru Mai,Ping Mai,Huashuo Mai and Fushuo Mai has increased, the proportion of Huan Mai,Xian Mai and Chenxixian Mai has unchanged(*p < 0.05).The score of specific endowment has significantly decreased(** p<0.01).After the second intervention of the control group,white and normal complexion increased,while red and light white complexion decreased(*p<0.05).4.More than 89.4% of the specific endowment constitution intestinal flora was composed of Firmicutes and Bacteroidetes,and the ratio between Firmicutes and Bacteroidetes was 28.9%.In addition,the composition of intestinal flora was significantly different among individuals.5.In experimental group,after the first intervention of Dog Days plaster,the abundance of Coprococcus₃ and Ruminococcaceae_UCG-014 has increased,while that of Fusobacterium has decreased(*P < 0.05).After the second intervention,the abundance of Faecalibacterium,Haemophilus and Barnesiella has increased(*p<0.05).In control group,after the second intervention,the abundance of Firmicutes has increased(*p < 0.05),the abundance of Faecalibacterium has significantly increased(**p<0.01),and the abundance of Bacteroides has decreased(*P < 0.05).Beside,Dog Days plaster intervention can reduce the difference of microflora between subjects.Conclusion: 1.The " winter disease being cured in summer " has a clear therapeutic effect on allergic rhinitis,cough variant asthma and classic asthma,and the combination of traditional Chinese medicine,Chinese patent medicine,western medicine and other treatment methods can improve the effective rate,prolong the treatment effect.As a result," winter disease being cured in summer " can be vigorously promoted in clinical practice.2.The Dog Days plaster can reduce the conversion score of specific endowment constitution,balance Yin and Yang in the specific endowment constitution,and achieve the effect of prevention before disease onset.3.The Dog Days plaster can change the composition of intestinal microflora of the specific endowment constitution people,which is one of the mechanism and theoretical basis of Dog Days plaster to play the role of "treating disease before
