Objective: To explore the effect of Biminkang Mixture on high secretion of allergic rhinitis(AR)by regulating the expression of MUC5 AC and MUC5 B in nasal mucosa under the guidance of Heli Yangqi Method.Methods: Based on the retrospective analysis of the research status of AR,it is proposed that "Heli Yang Qi Method " is the main treatment of AR.Biminkang Mixture can regulate the expression of MUC5 AC and MUC5 B in nasal mucosa and interfere with AR high secretion,which is verified by animal experiments.Sixty SD rats were randomly divided into six groups: blank group,model group,desloratadine tablet group and low,medium and high dose group of biminkang mixture,10 rats in each group.AR rat model was established and corresponding drugs were given for 14 days.Blood was taken from the abdominal aorta and nasal mucosa after treatment.The expressions of MUC5 AC and MUC5 B in blood were detected by ELISA.CD4 and CD8 expression in blood were detected by flow cytometry.Pathological changes of nasal mucosa were observed by HE staining.MUC5 AC and MUC5 B gene expression were detected by RT-PCR.The expression of MUC5 AC in nasal mucosa was detected by immunohistochemistry.Results:After the end of treatment,the behavioral evaluation score of each treatment group decreased.CD4 was decreased and CD8 was increased by flow cytometry,and MUC5 AC and MUC5 B concentrations were decreased by ELISA.Pathological observation showed that eosinophilic inflammatory infiltration was alleviated.The expression of MUC5 AC,MUC5B gene and MUC5 AC protein were decreased by RT-PCR and immunohistochemistry.There was significant difference between the test indexes and the model group(P < 0.01).Among them,the efficacy of biminkang mixture in medium andhigh dose group was better than that of diloratadine tablet group(P < 0.05).Conclusions:Nasal symptoms of AR rats were controlled by biminkang mixture.To improve the inflammatory infiltration of the nasal mucosa dominated by eosinophils.Decreased CD4 and increased CD8 in blood,The concentrations of MUC5 AC and MUC5 B were decreased.Decreased MUC5 AC,MUC5B gene and MUC5 AC protein expression in nasal mucosa.Then intervene in the high secretion state of AR to reduce the degree of AR lesions.It provides strong experimental evidence for the clinical application of biminkang mixture.
