| Objective: To reduce the recurrence rate is the difficulty and key point in the treatment of allergic rhinitis.A large amount of watery mucus is one of the main clinical manifestations of allergic rhinitis.Through the excavation of traditional Chinese medicine classical theory combined with years of clinical treatment experience,the tutor believes that "Yang qi loss in He Li" is the core mechanism of allergic rhinitis,and the traditional Chinese medicine intervention of allergic rhinitis should also focus on "Yang Qi".Binminkang mixture is a hospital preparation based on the above theory,mainly for the treatment of allergic rhinitis,the clinical effect has been proved in the early stage.Allergic rhinitis has the smallest persistent inflammation,which is also the key to the recurrence of allergic rhinitis.This expands our research idea of treating allergic rhinitis,and should not be confined to the symptoms of its onset.Studies have found that the mild persistent inflammatory state of allergic rhinitis can also be treated by Heyang Qi program.Nasal secretions also increased in AR patients with the most mild and persistent inflammation,mainly aquaporin and secreted mucin.Based on the role of Yang qi in water and liquid metabolism,the effects of Binminkang mixture on the expression of mouse nasal mucosa AQP1,AQP5,MUC5 AC and MUC5 B were tracked.To verify the effect of Binminkang mixture on the minimal persistent inflammation of allergic rhinitis.Methods: Female BALB/c mice with SPF grade were randomly divided into 5groups and given different treatments.Among them: MPI model group,MPI Western medicine group,MPI Chinese medicine group,blank group and AR model group.The experimental scheme designed in this paper is to construct a model of allergic rhinitis and mild persistent inflammation induced and stimulated by ovalbumin OVA.After completing the model construction,two treatment regimens of loratadine and Biminkang mixture were administered,and behavioral scores of the subjects were recorded.The nasal mucosa morphology of mice was observed and recorded by HE staining.The expressions of AQP1 and AQP5 in mouse nasal mucosa were detected by Western Blot.The expression of MUC5 AC and MUC5 B in nasal mucosa was observed by immunohistochemistry.RT-qPCR was used to detect the mRNA expression levels of AQP1,AQP5,MUC5 AC and MUC5 B in nasal mucosa.Results: The protein and mRNA expressions of AQP1,MUC5 AC and MUC5 B in nasal mucosa of mice in MPI model group were significantly increased(P < 0.01),the mRNA expression of AQP5 was decreased(P < 0.05),and the protein expression was significantly decreased(P < 0.01).After the intervention of Binminkang Mixture,the expression of AQP1,MUC5 AC,MUC5B protein and mRNA in nasal mucosa of MPI mice decreased(P < 0.05),the expression of AQP5 mRNA increased(P < 0.05),and the protein expression level increased significantly(P < 0.01).Conclusion: The traditional Chinese medicine preparation Biminkang mixture has the effect of regulating Yang qi in the body and improving the metabolism of water in the nasal cavity,so as to significantly improve the clinical symptoms of allergic rhinitis.Further observation shows that: After the intervention of MPI mice,inflammatory cell infiltration symptoms were alleviated,and the mRNA and protein expressions of AQP1,MUC5 AC and MUC5 B in nasal mucosa were decreased,while the mRNA and protein expressions of AQP5 were increased.In conclusion,Binminkang mixture can improve the structure of nasal mucosa on the basis of improving Yang qi,and has the function of repairing mucosa and reducing mucus exudation.It also has the therapeutic effect on MPI. |
