Aim:ApoE is the important component of plasma protein,which binds to the LDL receptor as ligan.It is involved in the transport of cholesterol in the blood,and regulates the metabolism of lipoprotein,which is also the main target of lipid-lowering drugs.Cerebral ischemic stroke is a general term of brain necrosis caused by the stenosis or occlusion of cerebral blood supply arteries(carotid and vertebral arteries)and cerebral blood insufficiency.Lipid metabolism plays an important role in the occurrence and development of ischemic stroke.There is a close relationship between ApoE gene polymorphism and blood lipid levels and ischemic stroke.This study is aimed to investigate the ApoE gene polymorphisms in healthy elderly people,hyperlipidemia people and stroke people,and to explore the relationship between ApoE gene polymorphism and hyperlipidemia,ischemic stroke in order to provide theoretical basis for the prevention and treatment of hyperlipidemia and ischemic stroke.Method:Primers were designed based on the gene sequences listed in the NCBI Gene database to construct an ApoE gene polymorphism detection kit.We have collected 324 patients with normal physical examination in the First Affiliated Hospital of Soochow University,376 patients with hyperlipidemia in the Department of Neurology,the First Affiliated Hospital of Suzhou University,and 342 patients with acute ischemic stroke admitted to the Department of Neurology,the First Affiliated Hospital of Suzhou University and detected The ApoE genotype by the constructed ApoE gene polymorphism kit.Gender,age,triglycerides,low density cholesterol and total cholesterol were collected in the three groups.Data were tested with SPSS 16.0 and Logistic regression analysis,and P<0.05 was statistically significant.Results:A total of 6 genotypes were detected in 324 healthy people with ApoE polymorphism detection system(ApoE?2/?2?ApoE?2/?3?ApoE?2/?4?ApoE?3/?3?ApoE?3/?4?ApoE?4/?4);5 genotypes were detected in 376 cases of hyperlipidemia and 342 cases of acute ischemic stroke(ApoE?2/?3?ApoE?2/?4?ApoE?3/?3?ApoE?3/?4?ApoE?4/?4).Due to the small number of samples,the statistical analysis of ApoE?2/?2 and ApoE?4/?4 could not be performed;The ApoE?2/?2,ApoE?2/?3,ApoE?2/?4 and ApoE?3/?3 genotypes in the normal human group were significantly different from those in the hyperlipidemia group and acute ischemic stroke(P<0.01,P<0.05).Three alleles(ApoE?2?ApoE?3?ApoE?4)were found in all three samples,in which ApoE?3 allele has the highest frequency of distribution;The frequency of ApoE?2 alleles in the normal human group was higher than that of the ApoE?4 allele;The frequency of ApoE?4 alleles in the hyperlipidemia group and acute ischemic stroke group was higher than that of the ApoE?2 allele.There was a significant difference(p<0.01)in the distribution of ApoE?2 and ApoE?3 alleles in the normal human group compared with the hyperlipidemia group and the ischemic stroke group.Analysis of acute ischemic stroke by logistic regression model found that hypertension,TC,TG,LDL-C were positively correlated with ischemic stroke.Conclusion:This experiment established a reliable ApoE gene polymorphism detection system;The ApoE?3 allele is the most widely distributed among the three studied groups;The frequency of ApoE?2 alleles in the normal human group was higher than that of the ApoE?4 allele;The frequency of ApoE?4 alleles in the hyperlipidemia group and acute ischemic stroke group was higher than that of the ApoE?2 allele.Regression analysis showed that ApoE?2 and ApoE?3 were protective factors for hyperlipidemia and ischemic stroke,while hypertension,TC,TG and LDL-C were risk factors for ischemic stroke.It also indicated ApoE gene polymorphism can provide a basis for early detection,prevention and treatment of ischemic stroke.
