Objective:By constructing a rat model of VCIND,to explore the effect of Bujingtiansuijiedu treatment on the learning and memory ability of VCIND rat model.Through the detection of blood rheology of VCIND rats,the effects of Bujingtiansuijiedu treatment on blood rheology were investigated,and the changes of LTP in the CA1 area of hippocampus of VCIND rats were detected,At the same time,the levels of synaptophysin,dopamine,norepinephrine and 5-HT in the hippocampus were detected to explore the effect of the Bujingtiansuijiedu treatment on neural reconstruction,synaptic replastic and neurotransmitters,and to investigate the expression level of BDNF and bFGF in VCIND rats and to explore the method of detoxification Effects of Bujingtiansuijiedu treatment on the expression of neurotrophic factors in rats.Methods:SPF Wistar rats were selected to construct VCIND rat model by middle cerebral artery embolism and reperfusion.All rats were divided into six groups:model control group,donepezil group,high dose group of traditional Chinese medicine,medium dose group of Chinese medicine,low dose group of traditional Chinese medicine,and sham operation control group.Then different intervention drugs were used to treat the rats in each experimental group for 5 days.The learning and memory ability of rats was detected by Morris water maze.The distance of escape latency,swimming distance in the platform quadrant,the memory frequency,the total distance of different quadrant swimming,the swimming time in the platform quadrant,the ratio of the platform quadrant to the total swimming time in the platform quadrant,and the number of times through the platform were measured respectively.The hemorrheology of rats after the treatment of Bujingtiansuijiedu treatment was carried out,including the local cerebral blood flow of the hippocampus,the viscosity of the whole blood(high-shear viscosity,medium-shear viscosity and low-shear viscosity),red cell aggregation index and rigidity index.The rat hippocampal slices were made to detect the changes of LTP in the hippocampal Schaffer lateral branch to CA1 pathway,and the levels of synaptophysin,dopamine,norepinephrine and 5-HT in hippocampus of rats were detected by ELISA.At the same time,qPCR and Western blot were used to detect the mRNA and protein expression levels of BDNF and bFGF in the hippocampus of rats.Result:Morris water maze experiment:compared with model group,Bujingtiansuijiedu method of traditional Chinese medicine can shorten VCIND rat escape latent period,increased in the platform quadrant swimming distance,frequency of memory,the total distance of different quadrant swimming,in the platform quadrant swimming time,platform quadrant and total swimming time frequency ratio,and the times of through the platform(P<0.05).Blood viscosity results:compared with model group,Bujingtiansuijiedu method of traditional Chinese medicine can reduce VCIND rats blood viscosity,including whole blood high-shear viscosity,whole blood viscosity,whole blood low-shear viscosity and plasma viscosity(P<0.05).Results of erythrocyte index:compared with the model group,Chinese medicine group can effectively reduce the erythrocyte pressure,erythrocyte aggregation index and rigidity index of VCIND rats(P<0.05).Changes in cerebral blood flow:compared with the model group,the changes of cerebral blood flow in VCIND rats can be effectively increased by using Chinese medicine(P<0.05).Changes of fEPSP at different time of electrophysiology in rats:compared with the model group,the changes of fEPSP in VCIND rats in 1 minute,30 minutes and 60 minutes were most affected by Chinese medicine(P<0.05).ELISA test results:compared with model group,Bujingtiansuijiedu method of traditional Chinese medicine can effectively increase the VCIND rats synapses in the:hippocampus,dopamine,norepinephrine and serotonin levels of expression(P<0.05).Expression of BDNF and bFGF:compared with the model group,the expression levels of BDNF mRNA,BDNF protein,bFGF mRNA and bFGF protein in hippocampal tissues of VCIND rats can be effectively increased by implantation and demulsification(P<0.05).Conclusion:the rat model of CIVDN was successfully constructed.Chinese medicine of Bujingtiansuijiedu method can effectively improve learning and memory ability and cognitive function in rats with CIVDN,and can effectively improve hemorrheology,reduce blood viscosity and increase cerebral blood flow in CVIDN rats.Bujingtiansuijiedu treatment can effectively promote the reconstruction of nerve and promote synaptic replastic in rats with CVIDN,and can also increase the expression level of neurotransmitters,effectively increase the expression level of BDNF and bFGF.
