| Objective:Adenomyosis,hereinafter shown as AM,is very common in the deaprtment of gynaecology,which severely hazards the patients'health in the clinical symptoms of secondary dysmenorrhea withprogressive aggravation,prolonged bleeding,overmuch menstrual blood,algolumegopareunia and infertility.The current known western medicines and operations for AM treatment have been proven with variety of limitations.The heart of the AM is blood stasis in Chinese medicine.The traditional Chinese medical science identifies phlegmatic hygrosis as one of the primary AM pathogenic factors,which causes blood stasis and ultimately leads to AM.Targeting at pelvic mass,this study develops "Chuanxia Ningkun decoction",and further explores its treatment effects on Symptoms and syndromes of adenomyosis in the macroscopic whole view and microscopic molecular biology view.From the viewpoint of micro molecular biology,this study considers adenomyosis ICR model mice as the subjects,takeing Survivin?VEGF?Caveolin-1 as entry points,passes through observing differences in model mice by three sets of indicators " Chuanxia Ningkun Decoction" in the treatment of the expression,investigats the Chuanxia Ningkun Decoction in the treatment of adenomyosis from different angles(phlegm and blood stasis)mechanism,Verifies efficacy?Method1.Animal ExprimentThis study considers adenomyosis ICR model mice as the subjects,in micro molecular biology Detectis contents of Survivin?VEGF?Caveolin-1?This study divids into two parts,animal model and animal experiment,lasts 6 months.1.1 Method1.1.1 Creat models:72 female ICR rats were randomly divided into 9 groups:model group 1,2,blank group 1,2,Chinese patent medicine control group(San Jie Zhen Tong)western medicine(Mifepristone),high dose group of Chinese Medicine,medium dose group of Chinese Medicine,low dose group of Chinese Medicine.There were eight mice in each group.Except blank group,all the others were fed on mixed withTamoxifen,Peanut oil,lecithin and refined milk for 4 days.Till 3 months,organizations were texted that wether myometrium appeared intimal invasion.Texted Survivin?VEGF contents in model group and blank group.1.1.2 Animal Expriment:Model group and blank group were fed on normal saline.Chinese Medicine groups were fed on "Chuanxia Ningkun Tang",Chinese patent medicine control group and western medicine group were fed on San Jie Zhen Tong and Mifepristone.Intragastric administration lasts 3 months.After 3 months,the mice were killed.The differences were texted in defferent groups by RT-PCR and immunohistochemistry.Objective to observe and analyze:Is there a difference in expressions of CAV-1 in model group and blank group?Is there difference in expressions of Survivin?VEGF?Caveolin-1 in different groups?2.Clinical researchThe clinical efficacy of "Chuanxia Ningkun Decoction" granules were defined by random observation,with "Chuanxia Ningkun Decoction" as experimental group,San Jie Zhen Tong as control group.2.1 Research method2.1.1 Through the comparative analysis between the experimental group and the control group,the curative effect in "Chuanxia Ningkun Decoction" relieved dysmenorrhea,reduce enstrual volume of AM(syndrome of phlegm stasis and blood stasis).2.1.2 Through the comparative analysis between the experimental group and the control group,the curative effect in "Chuanxia Ningkun Decoction" improved syndrome integration,CA125,uterine volume of AM(syndrome of phlegm stasis and blood stasis).Result1.Animal Expriment1.1 Modeling result:Model group and blank group were observed pathological sections.Model group can be seen that the muscular glands are hyperplasia afer tamoxifen drip irrigation for 4 days and normal feeding for 3 months.1.2 Experimental result:1.2.1 Survivin was highly expressed in AM mice.By treatment of high dose group of Chinese Medicine,expression of Survivin was decreased,but the expression in medium dose group and low dose group of Chinese Medicine were no depression.1.2.2 VEGF was highly expressed in AM mice.By treatment of high dose,medium dose and low dose groups,expression of VEGF all were decresed,and no significant difference among these groups.1.2.3 Caveolin-1 was highly expressed in AM mice.By treatment of high dose,medium dose and low dose groups,expression of Caveolin-1 were decresed.There were differences among different concentration groups.2.Clinical research2.1 After 3 months of treatment,NRS score both declined.The score of the experimental group was lower than that of the control group.It shown that"Chuanxia Ningkun Decoction"granules can improve the degree of dysmenorrhea,the curative effect is better than the Sanjiezhentong capsule.2.2 After 3 months of treatment,COX score both declined in two groups.The score of the experimental group was lower than that of the control group.It shown that"Chuanxia Ningkun Decoction"granules can improve dysmenorrhea,the curative effect is better than the Sanjiezhentong capsule.2.3 After 3 months of treatment,TCM syndrome integral declined in two groups,but there were no difference between the two groups.2.4 After 3 months of treatment,there were no difference in reducing uterine volume after treaments of experimental group and control group.It shown there is no obvious curative effect in improving the volume of uterus by "Chuanxia Ningkun Decoction".2.5 After 3 months of treatment,there were no difference in reducing CA125 after treaments of experimental group and control group.It shown there is no obvious curative effect in reducing CA125.2.6 After 3 months of treatment,MBL score both declined in two groups.Compared with control group,the effect is more significant in improving menstrual volume of experimental group.2.7 After 3 months of treatment,the effective rate was 84.84%in the experimental group,62.96%in the control group.It shown that "Chuanxia Ningkun Decoction" in the treatment of adenomyosis is always more efficient than San Jie Zhen Tong capsule.Conclusion1.Animal Expriment1.1 The method of mice model of adenomyosis by using tamoxifen for feeding neonatal mice is feasible.1.2 In utero in mice adenomyosis,Survivin and VEGF were highly expressed.After 3 months of treatment of "Chuanxia Ningkun Tang",expression of VEGF all were decresed,expression of Survivin was decresed only in high dose.It shows that "Chuanxia Ningkun ecoction" and may play a role in the treatment of adenomyosis by inhibiting angiogenesis and cell proliferation.1.3 The results showed that Caveolin-1 was highly expressed in the uterus of mice with adenomyosis.2.Clinical research2.1 "Chuanxia Ningkun Tang" can significantly improve dysmenorrhea symptoms,dysmenorrhea degree and duration of adenomyosis,and can effectively improve menstrual volume and TCM Syndromes.2.2 "Chuanxia Ningkun Tang"can neither reduce CA125 nor reduce uterine volume of patients with AM. |
