San Huang Decoction Improves Chronic Oxidative Stress And Inflammatory Microenvironment By Means Of Endocrine Therapy In Breast Cancer:Clinical And Experimental Study

Purpose:This study aims to observe the effects of San Huang decoction on the patients of hormone receptor positive breast cancer,including the improvement of symptoms,caused by endocrine therapy,to explore its mechanism from the angles of chronic oxidative stress,micro inflammatory reaction,blood stasis.we also explore the possible mechanism of San Huang decoction to anti migration and recurrence.Methods:62 patients with hormone receptor positive breast cancer were divided into two groups: treatment group has32 cases,besides endocrine therapy,they were provided San Huang decoction,but control group only use endocrine therapy.we recorded Kupperman score,SAS and SDS self-estimate score everymonth.Evaluate the improvement.:of oxidative stress syndroms,anxiety and depression index in treantmentgroup.Compare the changes of oxidative stress blood serum indexs,inflammatory reaction and blood stasis indexs in two groups,analyze the effect,safety and possible mechanism of San Huang Decoction on patients of breast cancer.At the same time,we use patients serum cultivated with MCF-7 and MCF-7/TAMR,The inhibition of MCF-7 and MCF-7/TAMR cells proliferation was determined by MTT assay.The expression of cell growth and apoptosis related proteins were determined by Western Blot analysis.Results:Because of long term endocrine therapy,the patients with breast cancer are always in a state of chronic oxidative stress and micro inflammation.they have the symptoms of similar menopausal or menopausal manifestations.Then influence their emotional and psychological changes.This study uses the Kuppermanscore,SAS and SDS self-estimate score to assess the patients life state.The research suggested the decline in the credits of insomnia,easy to be agited in treatment group.but in the control group,the score of Kupperman further went up.Compared with before into the group,the symptomatic improvement in the treatment group after 3 months including the manifestations of fatigue,hot flashes and sweating,headache,dizziness,waist and knee pain,dyspnea.The symptoms almost disappeared about similar menopausal and menopausal performance after six months treatment.While the total Kupperman score rised slightly in the control group when the experiment was over.this group patients always in a state of oxidative,emotional andpsychologicalstress.But the SDS and SAS indexs in experimental group declined gradually,have statistical defference(P<0.01 or P<0.05).This study seleced serum oxidative stress indicators(MDA.NO?SOD?GSH-Px),inflammatory indicators(IL-4?IL-6?TNF-a),blood stasis indexes(serum lipid parameter,hemorheological parameters,blood clotting,D-dimer),tumor prognosis targets(CA153?CEA?VEGF)as observed goal.patients in thetreatmentgroup after this study,the level of MDA and NO decreasd obviously than those before treatment.(P<0.05),while the indexs of GSH-Px and SOD were higher apparently than before treatment.(P<0.05)Compared with treatment group,those indicators no changes or even risedslightly.The levels of TNF-a and IL-6 decreasd than those before experiment(P<0.05),while IL-4 were distinctively higher than before treatment(P<0.01),the level of TNF-a rised slightly,but the level of IL-4?IL-6 as the same as before treatment.as far as blood stasis indexs were concerned,the level of TG?TC?LDL-C?apo-Bwere lower,while the level of HDL-C?apo-Ahigher than before.the indicators of TG?TC?HDL-C?apo-Ain the control group rised obviously than before treatment,ithas statistical difference.During experiment,the level of thetumor markersCA153 decreased than before,P<0.05,the level of CEA has no change after treatment,P>0.05.the level ofserum VEGF declined distinctly after 6 months treatment,P<0.05.Inthe cell experiment,itFurther tested San Huang Decoction could inhibit the proliferation of MCF-7 and MCF/TAMR.Itcan downregulated the protein expression of MAPK/ERK signal pathway.Conclusion:In conclusion,our findings indicated that San Huang decoction was able to influence the patients internal environment of hormone positive breast cancer,San Huang decoction performed to improve the stress symptoms,reduced the levels of oxidative stress,inflammatory factors,the indacators of blood stasis and CA153,CEA,VEGF,improved the life quality of the patients,reduced the side effects of hormone therapy.thus improving the prognosis of patients with breast cancer and prevent recurrence,metastasis and drug resistance.
