Objective:1.To observe the protective effects of moxibustion and acupuncture on gastric mucosa in rats with chronic atrophic gastritis?CAG?by replicating CAG rat models;2.To observe the effects of moxibustion and acupuncture on gastric mucosal protection related factors?EGF,TGF-?,PGE2?in CAG rats,and to explore the differences of moxibustion and acupuncture from the level of gastric mucosal protection related factors;3.To observe the effects of moxibustion and acupuncture on gastric tissue metabonomics in CAG rats using 1H NMR,and to explore the differences of response mechanism of moxibustion and acupuncture target organ effects from metabolite level.Methods:1.The CAG rat models were replicated by MNNG-ethanol gavage and abnormal of starvation and full method,which were divided into the normal group,model group,moxibustion group and acupuncture group.Rats in moxibustion group and acupuncture group were interfered with moxibustion and acupuncture at Zusanli point and Zhongwan point,and their general behavioral changes,weight changes and gastric tissue changes were observed while the histopathological changes of gastric tissue were observed by HE staining;2.The level changes of gastric mucosal protection related factors EGF,TGF-?and PGE2 were detected by IHC and ELISA;3.The changes of metabolites in gastric tissue were detected by 1H NMR.The metabolic patterns and differential metabolite analysis of gastric tissue were analyzed by PCA,PLS-DA and OPLS-DA.Results:1.After modeling,the gastric mucosa in the model rats changed in the following aspects:pale color,thinning layer,reduced elasticity,flattened folds,decreased mucus and scattered bleeding.After HE staining,the pathological examination of gastric tissue under light microscope showed a lot of inflammatory cell infiltration,a large number of major cell necrosis and vacuolar changes,a large number of fibrous tissue filling,various concentrated bleeding,glandular structure disorder and decrease,and inherent glandular atrophy.2.After moxibustion and acupuncture intervention,the mental state of rats improved,the fur lustred and the activity increased in moxibustion group and acupuncture group while the body weight showed steady and steady growth with no significant difference compared with the rats in the model group?P>0.05?.The effect of moxibustion and acupuncture on gastric histopathology showed that the pathological damage of gastric tissue in both groups was improved compared with that of the model group,and the difference was statistically significant?P<0.05?,but there was no significant difference between moxibustion group and acupuncture group?P>0.05?.3.The levels of EGF and TGF-?in gastric tissues of rats in moxibustion group and acupuncture group were significantly higher than those in model group?P<0.05,P<0.05?.There was no significant difference in the content of EGF and TGF-?between moxibustion group and acupuncture group?P>0.05?.The content of PGE2 in moxibustion group and acupuncture group was significantly higher than that in model group?P<0.01,P<0.05?,and the content of PGE2 in moxibustion group was significantly higher than that in acupuncture group?P<0.05?.4.From the PCA scores of each group,we can see that the metabolism of gastric tissue in CAG rats changed;moxibustion and acupuncture had an effect on the metabolism of gastric tissue in CAG rats and after moxibustion intervention,the metabolism of gastric tissue recovered significantly.OPLS-DA analysis showed that there were 17 differential metabolites in the gastric tissue of CAG rats,among which the concentrations of 7 metabolites were significantly increased:Lac,Gln,GSH,N-NAA,UDG,Ino and AMP while the concentrations of 10 metabolites were significantly decreased:Leu,Tau,DMG,Bet,Ser,PC,GPC,Ura,UD and Phe.The concentrations of the following metabolites reversed after moxibustion intervention:GSH,NAA,PC and Ura while there was no metabolites with concentration reversal detected after acupuncture intervention.Conclusion:1.MNNG-ethanol gavage and abnormal of starvation and full method can successfully replicate CAG rat models.2.Both moxibustion and acupuncture have gastric mucosal protection in CAG.3.The protective effect of moxibustion and acupuncture on the gastric mucosa of CAG may be related to the content changes of EGF,TGF-?and PGE2.The effect of moxibustion on PGE2 is more obvious.There is no significant difference in the impact of moxibustion and acupuncture on the changes of EGF and TGF-?level.4.Gastric mucosal cell membrane damage,mucosal cell apoptosis and neurotransmitter disorder were present in the gastric tissues of CAG rats,which had a certain impact on the rat nervous system,causing decreased body immunity accompanied by abnormal energy metabolism.There is difference in the effect of moxibustion and acupuncture on the metabolism of gastric tissue in CAG rats.Moxibustion is conducive to CAG rat gastric mucosal repair and immunity increase.However,the results of this experiment have not found differential metabolites associated with acupuncture treatment of CAG.It is possible that metabolic pathway of acupuncture is different from that of moxibustion,but the results need further analysis.
