Objective:1.To observe the effect of anti-blood stasis treatment on OHSS.2.To explore the mechanism of anti-blood stasis herbs in OHSS rat model of vascular permeability barrier.3.To screen efficient serum diagnostic markers of OHSS.Method:1.68 subjects were selected as subjects and divided into three groups. Patients suffered from OHSS were divided into two groups:Chinese medicine group was treated with anti-blood stasis herbs, aspirin group was given aspirin therapy. To check the ovulation rate, pregnancy rate, abandonment rate,the serum level of IL-6, VEGF and E2 in patients.2 To generate OHSS model of rat, the rats were divided into normal control group, OHSS group, aspirin group and low, middle and high dose group of Chinese medicine. To detect NO, VEGF, IL-6 levels, weight and ovarian weight of rats, then compare the differences.3. The serum of OHSS patients and healthy were subjected to mass spectrometry for proteins analysis, the proteins with significant difference were confirmed by western blot and enzyme linked immunosorbent assay.Result:1.There were lower periodic abandon rate and number of enlarged follicle in herbs group compared withaspirin group.2.The level of E2 and vascular endothelial growth factor (VEGF), abdominal perimeter, pelvic effusion of herbs group were significant lower than that of aspirin group.3. The level of P, VEGF, IL-6, EB levels of peritoneal and ovarian in OHSS group were significantly higher than the other group.4. The weight, ovary weight, ascites, levels of EB, NO, IL-6 and VEGF in herbs group were significantly lower than those in the aspirin group.5.57 candidate proteins were obtained from mass spectrometry screening.Compared with the control group, there were 29 proteins were up-regulated and 28 proteins were downregulated in OHSS group.6.The efficient and specific prediction of Haptoglobin, fibrinogen and lipoprotein lipase for OHSS was confirmed by ROC curve assay.Conclusion:The treatment of OHSS patients with the anti-blood stasis herbs can significantly reduce the level of inflammatory factors in patients and improve the ovulation rate.It can decrease abandonment rate without affecting the clinical pregnancy rate.2 It confirmed that anti-blood stasis herbs can significantly reduce inflammatory factors in OHSS rats, and has obvious protective effects on rat ovarian weight and ascites.3 Through mass spectrometry screening and further research validate that haptoglobin, fibrinogen, lipoprotein lipase can be used as serum diagnostic biomarkers forOHSS prognosis with high specificity and efficient.
