Effects Of Tonifying The Kidney Invigorating The Spleen Activating Blood Recipe On Integrin β3 And LIF Of Controlled Ovarian Hyper Stimulation Mouse Endometrial At The Implantation Window

ObjectiveTo observe the expression of integrinβ3 and leukaemia inhibitory factor(LIF) mRNA in endometrium of controlled ovarian hyper stimulation mouse at the implantation window, for investigating the effect of the recipe of tonifying the kidney invigorating the spleen activating blood on endometrial receptivity of controlled ovarian hyper stimulation mouse.MethodsHealthy and nonpregnant female KunMing mouse which had successive twice normal estrus cycles were randomly divided into 3 groups:30 cases of the traditional chinese medicine(TCM) group,30 cases of the aspirin group and 10 cases of the control group. Model of controlled ovarian hyper stimulation mouse were built. On the 3rd day of mouse estrus cycles, they were made by successive injection of alarelin acetate for 9 days. On the 9th day of injection of alarelin acetate, mouse were injected of pregnant mare serum gonadotrophin. After 48 hours of that, they were made by injection human chorionic gonadotropin. At the same time of model building, the mouse of 3 groups were given tonifying the kidney invigorating the spleen activating blood TCM, aspirin and 0.9% sodium chloride injection by gavage once a day for 11 days respectively. Mouse were sacrificed after 48 hours of intraperitoneal injection human chorionic gonadotropin, their middle segments of uterus were gotten for detecting expression of integrinβ3 and LIF messenger ribonucleic acid (mRNA) by PV two-steps immunohistological method and reverse transcriptase-polymerase chain reaction (RT-PCR). The statistical analysis methods of data:the count materials were evaluated with x2 test; the grade materials were evaluated with nonparametric test ridit analytical method; measurement data were analyzed by one-way analysis of variance analytical method.Results1.The protein expression of endometrial integrin (33 of controlled ovarian hyper stimulation mouse in the TCM and aspirin groups were statistical significantly higher than that in the control group(both P<0.01).while the expression of integrinβ3 in the TCM group were statistical significantly higher than that in the aspirin group (P<0.05).2.The endometrial expression of LIF mRNA of controlled ovarian hyper stimulation mouse in the TCM and aspirin group were statistical significantly higher than that in the control group(both P<0.01). while the endometrial expression of LIF mRNA in the TCM group were statistical significantly higher than that in the aspirin group(P<0.05).Conclusions1. Both of the recipe of tonifying the kidney invigorating the spleen activating blood and aspirin could increase expression of integrinβ3 and LIF mRNA of controlled ovarian hyper stimulation mouse endometrium at the implantation window.2. The recipe of tonifying the kidney invigorating the spleen activating blood may be more efficient than that of aspirin to improve the endometrial receptivity of controlled ovarian hyper stimulation mouse at the implantation window.
