Objective:To investigate the effect of Tianwang fuxin fang against myocardial ischemia and its possible mechanism.1. Myocardial ischemia model was induced by coronary artery ligation in rats, different interfers were continuous for21days, ST segment of ECG, myocardial infarct size, content of serum creatine kinase (CK), and lactate dehydrogenase (LDH), content of SOD and MDA in myocardial tissue were observed.2. Myocardial ischemia model was isoprenaline injectce repeatedly, the effects of Tianwang fuxin fang on myocardial damage degree and levels of creatine phosphokinase (CK) and lactate dehydrogenase (LDH) in serum.3. Normal dogs as the research object, the effect of Tianwang fuxin fang on animal blood pressure, heart rate, left ventricular pressure, pressure changing rate, aortic blood flow, blood flow of coronary artery, total peripheral resistance, myocardial oxygen consumption index by eight channel physiological recorder. 4. Rat model of blood stasis was induced by injection of adrenaline combinated with ice water, to observe the effect of Tianwang fuxin fang on blood rheology.5Study the of effect different doses of Tianwang fuxin fang on arterial thrombosis (arteries) time (OT) in rat carotid artery thrombosis by electrical stimulation.6. platelet rich plasma in normal rats were used,5-adenosine two phosphoric acid two sodium salt (ADP)was added, platelet aggregation in1minutes (PAG1),5min (PAG5) and the maximum peak (PAGM) were observed.7. Myocardial ischemia model in rats was induced by coronary artery ligation, different interfers were continuous for21days, the expression of TLR4-NF κ BP65in myocardial tissue were checked byimmunohistochemistry and PCR.1. Tianwang fuxin fang can significantly reduce the extent of myocardial injury and necrosis area, decreased content of creatine kinase (CK) and lactate dehydrogenase (LDH) in plasma, improved SOD activity in myocardial tissue, reduced the content of MDA. The results show on the Tianwang huxin fang has protecive effect on myocardial injury induced by myocardial ischemia animal. 2. Tianwang fuxin fang has obvious protective effect on sub acute myocardial ischemia induced by isoproterenol injection, reduced degree of myocardial injury, and inhibited the levels of creatine phosphokinase and lactate dehydrogenase (LDH) in serum.3. High and middle dose of Tianwang fuxin fang can slightly lower blood pressure, increase blood flow of coronary artery and aortic flow, reduce the total peripheral resistance, reduce myocardial oxygen consumption. But it hasnot significant effect on heart rate.4.Tianwang fuxin fang have obvious effect on whole blood viscosity under different shear rate in blood stasis rats model, and inhibit plasma viscosity.5. Tianwang huxin fang can obviously prolong carotid artery thrombus formation time in rats with electric stimulation, the result shows Tianwang fuxin fang can inhibit the formation of thrombosis.6. In1min,5min test point the effect of Tianwang fuxin fang on the rate of platelet aggregation was significantly decreased than that of the control group, the platelet aggregation peak was significantly lower than the control group, that suggested that Tianwang fuxin fang can inhibited platelet aggregation significantly.7. Tianwang fuxin fang has inhibitory effect on expression of the TLR4protein and gene, NF-κB p65protein in myocardial ischemia rats. Conclusion:Tianwang fuxin fang has the effective with anticoagulant, anti thrombosis and myocardial ischemia, and its mechanism may be related to modulating the inflammatory signal TLR4-NF κ B.
