BackgroundReview the traditional and modern TCM' s research on the name, the cause, the mechanism, the diagnosis, the syndrome, the therapy , the prevention of the disease such as "the Benign prostatic hyperplasia(brief BPH)". Summarize the origin and development of the treatment of "Jing Long" by "tonyfying the kidney and reinforcing the spleen to remove stasis ". Review recent years' condition about TCM and the combining of TCM and western Medicine' s clinic treatment on BPH, and the condition about TCM' s experiment research about BHP. Summarize the relationship between older' s kidney dificiency and calcium' s overload inside cell of and the relativity research about the outbreak of Atherosclerosis obliterans(ASO).Form the expectation about the relation of the outbreak of BPH and calcium' s overload inside cell by the principle of"treating difference disease with the same method", carve out TCM' s new research method on BPH.purposeChoose male SD rats and repeat the model of Benign prostatic hyperplasia rat by popular method. Use the effective prescription of "tonyfying the kidney and reinforcing the spleen to remove stasis "(brief YST),which is used by my tutor professor Cui Xuejiao to treat BPH in clinic, to do medical intervene. Set nomal group , model group, treatment group and comparison group and experiment indexes. Observe the effection of YST on rat' s benign prostatic hyperplasia and the influence of YST on calcium' s overload inside rat' s benign prostatic hyperplasia cell. Discuss the relationship between calcium' s overload inside cell and the origin and development of the BPH. Explore more the mechanism of YST' s prevention and treatment on rat' s benign prostatic hyperplasia. Help exploiting new way and new medicine. Method1 experiment1.1 methodAccording to the random comparing principle, set the nomal group, model group and ZGLS comparing group,YST low, middle,high dose treatment groups, 6 rats in each group, 36 rats in total. Injecting testosterone propionate into castrated SD male rats to repeat benign prostatic hyperplasia model. Except the nomal group, other groups inject testosterone propionate into castrated SD male rats by 4mg/kg, go on for 30 days. In the 5 castrated rats groups, the 4 groups were fed with medicine except the model group.The comparison group was fed with ZGLS through stomach, and the treatment groups were fed with low, middle, high dose through stomach.The dose was calculated by the avoirdupois of clinical dose, that is ,7 times the human dose. ZGLS capsule group was fed by 0. 7g/kg, YST' s low, middle, high dose groups were fed by 0. 25g/kg,0.50g/kg, 1.00g/kg separately. Each medicine was mixed by 3ml distilled water to push into stomach one time each day. Normal group and model group were pushed with same cubage distilled water one time each day. During the period , the rats were separated by group, they can eat and drink freely. Sampling was on the 31st day after feeding medicine by exsanguinating and getting the prostatic of all groups' rats.1.2 index①weigh the prostatic of the rats in all groups and calculate the index ofprostatic;②make the pathology slices of the prostatic of the rats in all groups (HEdying);③test the calcium' s density in all rats' serum;④Immunohistochemistry immunity method to test the express of Fas' s and PCNAin rat' s prostatic hyperplasia cell;⑤Flow cytometer test calcium inside rat' s prostatic hyperplasia cell ineach groups;⑥research the ralativation among all indexes;1.3 result1.3.1 The rats' benign prostatic hyperplasia model was successful. The weight of the prostate and the index showed that the normal group and the model group have great dirrerency (P<0. 05),and the modalities under microscope have great difference;1.3.2 YST can effectively reduce the weight of rats' prostate (P< 0.05),which showed that YST have different restrain effect on the rat' s prostatic hyperplasia cell in dose groups;1.3.3 The result proved that YST' s low, middle, high dose groups and ZGLS comparison group' s Fas express are all higher than model group, and the express rise when the dose rise. The result also proved that YST' s middle and high dose group' s PCNA' s express are far less than the model group, and the dose in YST groups is negative related with PCNA' s express;1. 3. 4 About the index of calcium in prostatic cell, YST high dose group have distinct difference comparing with model group (P<0. 05).The calcium inside the cell reduced when the dose of YST increased by analyzing the average, which showed that YST may effectively restrain the overload of calcium inside rat' s prostatic hyperplasia cell. Non-linearity relation analysis proved that the calcium inside cell and the calcium in serum has no relation. ConclusionYST has great effect on rat' s benign prostatic hyperplasia.One of the mechanism is that it most properly reduce the overload of calcium inside prostatic cell, so that it can influence the express of Fas (which can promote cell' s death) and PCNA(which can promote hyperplasia), the former is improved and the later is reduced, and finally promote the prostatic cell' s death to restrain the prostatic hyperplasia.2 clinic researchBPH is a normal disease in oldman. It improves every year with the speeding of our society' s aging progress. On the other hand, the incident age is reducing in recent years, the middle-age sufferer is becoming more and more. This disease has greatly influenced the quality of them. So the prevention of BPH is getting more and more attention. The research is trying to discuss if the BPH of insufficiency of kidney-YANG and insufficiency of kidney-YIN have relationship with the density of calcium in serum and the weight of prostate.The author reviewed 102 cases of BPH in hospital .He try to expand the meaning of TCM syndrome differentiation and serve the clinic better.2.1 methodAccording to the BPH diagnose standard and the syndrome differentiation , bring into 102 BPH cases of insufficiency of kidney-YANG and insufficiency of kidney-YIN in hospital , each syndrome has 51 cases. The age is beween 51 and 87, average 68. Among these, 22 cases are beween 51 and 60, 49 cases are beween 61 and 70, 27 cases are beween 71 and 80, 4 cases are over 81. The course of diseases is from 3 months to 23 years. The index were the density of calcium in serum and the weight of prostate. The frondose method is to collect the density of calcium in serum on the next morning in the hospital and alse the bulk data of each cases' prostate from the result of type-B ultrasonic. According to the normal proportion 1.00~1.05, calculate the weight of prostate(g)=length(cm)×width(cm)×thickness(cm)×0. 57×1. 05g/cm3。2.2 result2.1 Comparing the calcium in serum , the syndrome of insufficiency of kidney-YANG and damp-heat have no distinct difference by statistics (P>0. 05);2. 2 Comparing the weight of prostate, the syndrome of insufficiency of kidney-YANG and damp-heat have no distinct difference by statistics;2.3 Analysing the non-linearity relativity between the syndrome of insufficiency of kidney-YANG and damp-heat on calcium in serum and the weight of prostate, they have no distinct difference by statistics (P>0. 05), the coefficient are both under 0.3, which hint that the two indexes have no relativity.ConclusionThe result of this research matches the result of animal experiment by the same indexes. But the comparison of the relativity between the calcium inside cell and the weight of prostate showed that these two indexes have relativity in the animal experiment. Thus we can dope out that : BPH sufferer' s calcium in cell has relativity with the weight of prostate, we should develop research in this direct.
