Allergic rhinitis is a common disease, it can take place in all ages. But, it is often seen in teenage. Allergic rhinitis can occur in all year, nevertheless;it is more ofter seen in fall and winter. Especially, when the change of weather or the stimulaion of peculiar smell and air pollutant can induce allergic rhinitis. The last twenty years, the incidence, of allergic rhinitis has a remarkable tendency of increase.The statistics of incidence is 2% to 20%, the average is 10% in America. The average is 37.74% in China.Allergic rhinitis belongs to type 1-hypersensitivity, the pathological? change was on the epithelium of nasal mucosa, base membrane and gland, we can find the dilation of blood capillary and the increase of permeability, these will cause edema of mucosa.At the same time, we can find infiltration of play mor phonuclear-eosinophil. The symptoms of allergic rhinitis are nasal itch, sneeze, nasal congestion, rhi norrhea. The characteristic of allergic rhinitis are paroxysmal and immediate and break out again and again, also the course of disease is long, and it is difficult to cure. Allergic rhinitis brings much pain to patient, it can affect the patient's work and stady.It is more important to find out the method to take away symtoms of allergy, to reduce the recurrence, to shorten course of desease, to elevate the effect of remedy. According to the nidus of allergic rhinitis is at nose. As a result, we use external theapy is a good method to treat the disease.If we use a special manufacture to make traditional Chinese medicine become smaller particle, it can pass through the gap of nasal mucose to blood vessel, for the purpose of blood circulation can produce the fast effect of therapy. We use Nanotechnology can make this ideal come true. This purpose of this research is use AR-rat-model to examine the effect of therapy and pathology of nanotechnological traditional Chinese medicine. At the same time, we also test the quality and quantity of nanotechnological traditional Chinese medicine.Method of experimentranimal-rat, SD. make AR-rat-model: Male, SD, rat had feed for five days. Body-weigh of rat are between 180-200 gm. TDI is dissolved in olive oil toallocate 10% TDI-olive-solution. We use pippet to suck lQal solution, drop into the rat's nasal cavities. After evaluation, the total score of nasal symptom >5, and, there are a lot of polymorphonuclear eosinophils in nasal secretion. It shows the mode is successful, then, groups into complete-blank control group (unmodel), model group (no drug), positive control group (Ditong biyan shui group), test group (shubi penji group).The test groups are separated three groups from different dosage, large dose group, middle dose group and small dose group. Result: 1 The effect of AR-rat model's symptom of nose and WBC in secreation.?We compare with model group, the shubi penji group and Ditong biyan-shui group both has the most remarkable reduce effect in sneeze's score, itch's score, rhinorrhea's score, and score of symptom. It shows as to rhinitis symptom's total score has remarkable reduction, (P<0.01).?The shubi penji group and Ditong biyan shui group have a remarkable reduction in the amount of polymorphonuclear eosinophil of nasal-secretion. (P<0.01)o 2.The effect of AR-rat model's index of 1 blood biochemistry comparing with model group, shubi penji group and Ditong biyan shuigroup both have reduce the level of IL-2 and IL-4;Middle dose group can remarkably decrease plasma IL-6 (P<0.05)o small dose group, large dose group and ditong biyan shui group all can decrease the level of plasma IL-6° Large dose group, middle dose group and ditong bi yan shui group all can remarkably decrease the level of plasma SP (P<0.01)o Small dose group can obriously reduce the leve of plasma SP (P<0.05)o Large dose group, middle dose group and ditong bi yan shui group all can remarkably raise the level of plasma SOD (P<0.01)o small dose group can goup the level of plasma SOD.3.AR-rat-model's pathologic hange of nasal mucosa comparing with model group, large dose group, middle dose group and ditong bi yan shui group have remarkable reduction of pathologic change of nasal mucosa. (P<0.01), small dose group has obviously reduction of pathologic change (P<0.05)o 4.Effect of AR-rat-model's ultramicro-struction of nasal mucosa?Comparing with model group, large dose group, middle dose group and di tong bi yan shui group all can remarkably raise density of cilium of nasal epithelium (P<0.01)<,?Comparing wiht model group, large dose group, middle dose group small dose-group all can remarkably increase the density of mitochondria (P<0.01)o Large does group and small dose group can obviously decrease specific surface superficial measure (P<0.05)o Middle dose group can remakably decrease specific surface superficial measure (P<0.05)<> Large dose group, middle dose group and small dose group group all can remakably raise specific membrame superficial measure (P<0.01)o It shows shu-bi-penji can remarkable go up metabolism of cell so that can improve situation of allergic rhinitis. 5.Effect of the expression of AR-rat-model's bcl-2m RNA, bcl-2 and bax?Comparing with model group, large dose group can remarkably decrease the pu score of bcl-2 mRNA (P<0.01), middle dose group can obviously decrease the pu score of bcl-2MRNA(P<0.05)o?Comparing with model group, large dose group can middle dose group can remarkably decrease pu score of bcl-2 (p<0.01)> large dose group can remarkably increase pu score of bax (P<0.01) o It shows Shu-bi-penji can promote decease of inflammation cell and multiplication cell in nasal mucosa. 6.Aute toxic experiment of shu-bi-penjiAnimal:rat. SD male, experimental dosage:three hundred times of clinical dosage period of experiment: three times/day reesult: It shows experimental rats have normal activity and nasal mucosa are normal too. The experiment proves shu-bi-penji is safe. 7.Determination of mass standard of shu-bi-penjiAfter using HPLC and comparing with standard sample, it shows nano technological traditional Chinese medicine has lost about 39% of main ingredient.Conclusion:?Shu-bi-penji can buffer nasal symptoms of AR-rat-model agrees with result of pathological section of mucosa.?Shu-bi-penji can reduce the amount of poly morphonu clear-eosinophil in nasal secretiion agrees with examination of blood biochemistry index.?Shu-bi-penji can repair nasal mucosa tissue of AR-rat-model agress with examination of nasal mucosa's ultra structure checked by electromicroscope.?Shu-bi-penji can adjust the unblance of control gene again agress with the method ofhybrid procedure and moleculobiological.
